Immunomodulatory effects of flavonoids: An experimental study on natural-killer-cell-mediated cytotoxicity against lung cancer and cytotoxic granule secretion profile

Background: A new approach involving immune-cell-mediated cancer therapy has been adopted extensively for the sake of lung cancer treatments by utilizing natural killer (NK) cells. NK cell activity can be enhanced with certain agents, and among them are flavonoids. Thus, this study was conducted to investigate the immunomodulatory roles of apigenin, luteolin and quercetin on NK cell activity against lung cancer cells and on the secretions of perforin and granulysin profile. Methods: The NK-92 cells were grown in complete alpha-Minimum Essential Medium (MEM). NCI-H460 lung cancer cells were cultured in Roswell Park Memorial Institute 1640 media. NK cell activity against lung cancer cells were done using MTT(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. The secretions of perforin and granulysin profiles were then analysed using enzyme-linked immunosorbent assay. Results: Apigenin, luteolin and quercetin significantly increased the NK-cell-mediated cytotoxic activity against lung cancer cells at concentrations 12.5 mu g/ml and 25 mu g/ml (P < 0.001). The secretion levels of perforin and granulysin from NK cells were also significantly enhanced with apigenin and luteolin treatment but not with quercetin. Conclusions: All three flavonoid compounds possessed some significant immunomodulatory actions on NK cell cytotoxic activity and granule secretion profiles towards lung cancer therapy. MgFeAl layered double hydroxide prepared from recycled industrial solid wastes for CO2 fixation by cycloaddition to epoxides

MgFeAl trimetallic layered double hydroxide (LDH) was prepared using industrial solid wastes of red mud and ferronickel slag, and exfoliation of the LDH by acetone washing resulted in a final product with high surface area (319 m(2)/g). It showed a CO2 adsorption capacity of 70.2 mg/g at 298 K and 1 bar, which was somewhat higher than that of a similar LDH synthesized using pure chemicals (63.8 mg/g). The LDH was tested as a catalyst for solvent-free CO2 cycloaddition to various epoxides. Significant conversion of epoxides and high selectivity to the corresponding cyclic carbonates were obtained under mild reaction conditions. The LDH achieved an excellent result of 98.0% conversion of epichlorohydrin and 98.2% selectivity to the cyclic carbonate in the presence of tetrabutylammonium bromide as co-catalyst at 50 degrees C and 5 bar after 7 h, catalyzed by the combination of Lewis acidic metal sites and bromide ions. The LDH was recovered after the reaction and maintained good catalytic performances after five repeated runs without any significant loss in catalytic activity. This work demonstrated that effective CO2 capture/utilization could be achieved simultaneously using a porous material easily prepared from industrial wastes.

Influence of Lithium Bromide on Electrical Properties in Bio-based Polymer Electrolytes

This research presents the influence of lithium bromide (LiBr) on the electrical properties of alginate in bio-based polymer electrolytes (BBPEs) system. Bio-based alginate was prepared using the solution casting technique with various LiBr compositions. The ionic conductivity and electrical properties of the prepared BBPEs samples were studied using electrical impedance spectroscopy over a frequency range of 50 Hz-1 MHz. A maximum ionic conductivity of 7.46 x 10(-5) S cm(-1) was obtained for a sample containing 15 wt. % lithium bromide-doped alginate BBPEs at ambient temperature (303 K). The electrical analysis revealed that the most conductive sample based on alginate-LiBr BBPEs has optimum dielectric constant and loss, which significantly increases as temperature increases. The dielectric properties show that all alginate-LiBr BBPEs are in non-Debye behavior conditions, where no single relaxation occurs in the present system.

Repellence and acute toxicity of a nano-emulsion of sweet orange essential oil toward two major stored grain insect pests

Control strategies in stored-product facilities mainly rely on synthetic pesticides. The development of new environmentally friendly alternatives, such as essential oils (EOs), is a key issue. The aim of this research was to develop a stable nano-emulsion containing a high amount of sweet orange (Citrus sinensis) EO and to evaluate its insecticidal activity against Tribolium confusum du Val (Coloptera: Tenebrionidae) and Cryptolestes ferrugineus (Stephens) (Coleoptera: Cucujidae). The experimental nano-emulsion showed a low surface charge (zeta slightly higher than -30 mV) and a droplet size within the sub-micrometre range (131.37 nm +/- 0.29), maintained also after 1 year of storage. The EO nano-emulsion presented good repellence toward both adult insects, which lasted until 24 h of exposure at the highest dosages. Furthermore, the developed nano-formulation showed acute toxicity against both insects when tested as fumigant and cold aerosol. In fumigation trials, adults of C. ferrugineus were more susceptible than T. confusum adults. Moreover, the insecticidal activity of cold aerosol was 5 and 7-fold higher than fumigation against both C. ferrugineus and T. confusum, respectively. The EO-nano-emulsion was effective in controlling and repelling the target pests. Cold aerosol treatments with EO nano-formulations is a promising alternative method for the sanitation of production areas, warehouses, and machineries.

Detection of Tumor Marker Using ZnO@Reduced Graphene Oxide Decorated with Alkaline Phosphatase-Labeled Magnetic Beads

The magnetic-bead-based electrochemical enzyme-linked immunoassay (MB-eElisa) represents an attractive approach to develop cost-effective systems that are suitable for sensing complex biological samples. Its sensitivity essentially depends on the transduction efficiency of enzyme catalytic reactions into electrochemical responses. Here, an ultrahigh signal-to-noise alkaline phosphatase (ALP)-based MB-eElisa system is developed with a renewable zinc oxide-reduced graphene oxide composite modified carbon paste electrode (ZnO@rGO/CPE). This biosensing system employs one antibody decorated MB (MB-Ab(1)) to capture a model tumor marker-carcinoembryonic antigen (CEA)-from samples, while other antibody coated gold nanoparticles-ALP bioconjugates (Ab(2)-AuNPs-ALP) convert 1-naphthyl phosphate (1-NPP) into electroactive 1-naphthol (1-NP). Benefitting from the unique electrochemical properties of a ZnO@rGO/CPE, including nearly zero background and significantly enhanced responses toward the hydrolyzed 1-NP in the presence of trace surfactants, the MB-eElisa system detects selectively CEA in a calibration range of 0.01-6.0 ng mL(-1) and with a detection limit of 4.0 pg mL(-1) (S/N = 3). Such a system was further applied to the detection of CEA in serum samples of cancer patients. The combination of MB-based ALP-linked immunoassay with a ZnO@rGO/CPE thus establishes a reusable and inexpensive electrochemical sensing platform for the rapid and sensitive detection of ultratrace biomarkers in complex biological samples.

A novel oncolytic adenovirus inhibits hepatocellular carcinoma growth

Objective To evaluate the inhibitory role of a novel oncolytic adenovirus (OA), GP73-SphK1sR-Ad5, on the growth of hepatocellular carcinoma (HCC). Methods GP73-SphK1sR-Ad5 was constructed by integrating Golgi protein 73 (GP73) promoter and sphingosine kinase 1 (SphK1)-short hairpin RNA (shRNA) into adenovirus serotype 5 (Ad5), and transfecting into HCC Huh7 cells and normal human liver HL-7702 cells. The expression of SphK1 and adenovirus early region 1 (E1A) was detected by quantitative real-time PCR (qRT-PCR) and western blot, respectively. Cell viability was detected by methylthiazolyldiphenyl-tetrazolium bromide (MTT) assay, and apoptotic rate was determined by flow cytometry. An Huh7 xenograft model was established in mice injected intratumorally with GP73-SphK1sR-Ad5. Twenty days after injection, the tumor volume and weight, and the survival time of the mice were recorded. The histopathological changes in tumor tissues were observed by hematoxylin-eosin (HE) staining and transmission electron microscopy (TEM). Results Transfection of GP73-SphK1sR-Ad5 significantly upregulated E1A and downregulated SphK1 in Huh7 cells, but not in HL7702 cells. GP73-SphK1sR-Ad5 transfection significantly decreased the viability and increased the apoptotic rate of Huh7 cells, but had no effect on HL7702 cells. Intratumoral injection of GP73-SphK1sR-Ad5 into the Huh7 xenograft mouse model significantly decreased tumor volume and weight, and prolonged survival time. It also significantly decreased the tumor infiltration area and blood vessel density, and increased the percentages of cells with nucleus deformation and cells with condensed chromatin in tumor tissues. Conclusions GP73-SphK1sR-Ad5 serves as a novel OA and can inhibit HCC progression with high specificity and efficacy.
