Long non-coding RNA OIP5-AS1 (lncRNA OIP5-AS1) and microRNA-128-3p (miRNA-128-3p) have been reported to play significant roles in human diseases. However, their role in atherosclerosis (AS) has been less studied. The aim of this research was to reveal the roles and functional mechanisms of OIP5-AS1 and miRNA-128-3p in AS development. Quantitative real-time polymerase chain reaction (qRT-PCR) and western blot assays were performed to detect gene expression. Cell proliferation and apoptosis were assessed using a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay and flow cytometry analysis, respectively. In addition, ELISA was employed to determine the levels of interleukin (IL)-6, IL-1 beta, and tumor necrosis factor (TNF)-alpha. Oxidative stress was examined using a relevant kit. Furthermore, the interaction between miR-128-3p and OIP5-AS1 or cyclin-dependent kinase inhibitor 2A (CDKN2A) was predicted using StarBase, and then confirmed using the dual-luciferase reporter assay or the RNA immunoprecipitation (RIP) assay. We found that OIP5-AS1 and CDKN2A levels were upregulated and the miR-128-3p level was downregulated in oxidized low-density lipoprotein (ox-LDL)-induced THP1 cells. OIP5-AS1 knockdown weakened the regulatory effect of ox-LDL on cell progression. Interestingly, OIP5-AS1 directly interacted with miR-128-3p and miR-128-3p-targeted CDKN2A. Furthermore, OIP5-AS1 regulated ox-LDL-induced cell progression through modulating miR-128-3p expression, and miR-128-3p exerted its influence by modulating the CDKN2A level. Finally, we confirmed that OIP5-AS1 suppressed miR-128-3p expression to increase the level of CDKN2A. In conclusion, our findings demonstrate that OIP5-AS1 knockdown repressed the effect of ox-LDL on cell progression through regulating the miR-128-3p/CDKN2A axis, providing a potential target for the treatment of AS.

Preparation and characterization of positively surface charged zinc oxide nanoparticles against bacterial pathogens

Solvothermal synthesis was used to investigate the formation of zinc oxide (ZnO) nanoparticles (NPs). A series of ZnO NPs was synthesized with different relative ratios of didodecyldimethylammonium bromide (DDAB) and zinc nitrate (ZN). The variation in the molarity influenced the crystallinity, size, and morphology of the obtained ZnO NPs. X-ray diffraction, Fourier-transform infrared spectroscopy, field-emission scanning electron microscopy, high-resolution transmission electron microscopy, and zeta potential analysis were used to study the characteristic features of the ZnO NPs. The ZnO surface charge, size, and morphological structure were highly reliant on the concentrations of DDAB and ZN. With increasing relative ratio of DDAB to ZN, the particle size of ZnO NPs decreased and the surface charge increased to higher positive value. The ZnO NPs synthesized with cationic liquid DDAB presented enhanced performance in preventing the growth of Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli) strains. The antibacterial activity of ZnO NPs have direct contact with the microbial cell wall resulting in destruction of bacterial cell integrity, release of antimicrobial Zn2+ ions, and induce cell death. This is due to the positively charged smaller ZnO NPs, prepared with DDAB cationic surfactant, effectively acting as an antimicrobial agent against food-borne pathogenic bacteria.

Self-Limited Grafting of Sub-Monolayers via Diels-Alder Reaction on Glassy Carbon Electrodes: An Electrochemical Insight

The grafting of molecular monolayers is critical for the functionalization of surfaces. In molecular electrochemistry, the surface modification of electrodes and the way molecules are attached to the electrode surface are highly critical to electron transfers and electrochemical reactions. In this paper, sub-monolayers were covalently grafted onto glassy carbon (GC) electrodes via Diels-Alder cycloaddition with two soluble dienophiles, that is, propargyl bromide and ethynyl ferrocene. Such an approach is clean (no by-product, no catalyst/additive) and occurs under mild conditions by heating at 50 degrees C in toluene for few hours. The as-modified electrodes were thoroughly characterized by FTIR, XPS, and cyclic voltammetry using both millimetric GC electrodes and ultra-microelectrodes. Cyclic voltammetry gave access to surface coverage and clearly evidenced the covalent grafting of sub-monolayers. The grafting of functional sub-monolayers via Diels- Alder cycloaddition could be easily extended to various functionalities and carbons to prepare electrochemical sensors or electrocatalytic surfaces.

Porous nanosheets of carbon-conjugated graphitic carbon nitride for the oxidation of H2S to elemental sulfur

Graphitic carbon nitride (CN) has been reported as an emerging non-metallic catalyst for the oxidation of H2S to elemental sulfur, but its industrial application is hampered by the unsatisfactory catalytic activity. Herein, we report for the first time a facile bottom-up approach to synthesize porous nanosheets of in-plane carbon-conjugated CN (PCNUC) using cetyltrimethylammonium bromide (CTAB) and urea as precursors. The resulted PCNUC catalysts are porous and high in specific surface area because CTAB acts not only as a carbon source for conjugation with CN for the extension of pi-electron system, but also releases gases to function as soft template to create porous CN-based sheets. Thus, the carbon-conjugated CN displays high performance for the catalytic oxidation of H2S to S. Furthermore, the optimized PCNUC catalyst exhibits better durability than the traditional carbon-based and Fe2O3-based catalysts. The results of the present study open up a new avenue for the design of efficient CN-based catalysts for desulfurization reactions. (C) 2019 Published by Elsevier Ltd.

A BMSCs-laden quercetin/duck’s feet collagen/hydroxyapatite sponge for enhanced bone regeneration

Treating critical-sized bone defects is an important issue in the field of tissue engineering and bone regeneration. From the various biomaterials for bone regeneration, collagen is an important and widely used biomaterial in biomedical applications, hence, it has numerous attractive properties including biocompatibility, hyper elastic behavior, prominent mechanical properties, support cell adhesion, proliferation, and biodegradability. In the present study, collagen was extracted from duck’s feet (DC) as a new collagen source and combined with quercetin (Qtn), a type of flavonoids found in apple and onions and has been reported to affect the bone metabolism, for increasing osteogenic differentiation. Further, improving osteoconductive properties of the scaffold hydroxyapatite (HAp) a biodegradable material was used. We prepared 0, 25, 50, and 100 mu M Qtn/DC/HAp sponges using Qtn, DC, and HAp. Their physiochemical characteristics were evaluated using scanning electron microscopy, compressive strength, porosity, and Fourier transform infrared spectroscopy. To assess the effect of Qtn on osteogenic differentiation, we cultured bone marrow mesenchymal stem cells on the sponges and evaluated by alkaline phosphatase, 3-4-2, 5-diphenyl tetrazolium bromide assay, and real-time polymerase chain reaction. Additionally, they were studied implanting in rat, analyzed through Micro-CT and histological staining. From our in vitro and in vivo results, we found that Qtn has an effect on bone regeneration. Among the different experimental groups, 25 mu M Qtn/DC/HAp sponge was found to be highly increased in cell proliferation and osteogenic differentiation compared with other groups. Therefore, 25 mu M Qtn/DC/HAp sponge can be used as an alternative biomaterial for bone regeneration in critical situations.

Cytoprotective effect of crocin and trans-resveratrol on photodamaged primary human retinal pigment epithelial cells

Purpose: Light-induced damage to retinal pigment epithelium during pars plana vitrectomy remains a hot topic in ophthalmology. Improvements in technology led to a change of light sources, selective filters, and shorter light exposure time. Currently, there is no satisfying solution to the problem. The aim of the study was to investigate the cytoprotective effects of crocin and resveratrol on light-induced damage to primary human retinal pigment epithelial cells in vitro. Methods: Primary human retinal pigment epithelial cells were exposed to light analogous to the illumination during pars plana vitrectomy. To evaluate the cytoprotective effects and potential toxicity of resveratrol and crocin, human retinal pigment epithelial cells were incubated with varying concentrations of both before 3-[4,5-dimethylthiazol-2-yl] tetrazolium bromide (MTT) viability assay. Furthermore, glutathione levels were measured to investigate synergistic antioxidant potential. Apoptosis of human retinal pigment epithelial cells was determined by a nucleosome detection enzyme-linked immunosorbent assay. Results: Crocin and resveratrol improved cell viability in photodamaged human retinal pigment epithelial cells significantly from 40.65 +/- 21.99% in illuminated human retinal pigment epithelial cells and reached a peak viability of 85.64 +/- 11.37% in crocin and resveratrol pretreated cells (for all: p < 0.001). In line, the combination of the supplements increased glutathione levels significantly from 39.35 +/- 21.96% to 80.74 +/- 10.32% (p = 0.017). No toxic effects were detected (p > 0.99). However, no change in apoptosis rates could be observed following pretreatment with crocin and resveratrol (p > 0.99). Conclusion: Crocin and trans-resveratrol revealed cytoprotective effects on human retinal pigment epithelial cells supporting both supplement’s development as potential perioperative treatments in light-induced retinal pigment epithelial damage.

Magnetic methylene-based mesoporous organosilica composite-supported IL/Pd: a powerful and highly recoverable catalyst for oxidative coupling of phenols and naphthols

A novel magnetic methylene-based mesoporous organosilica composite-supported IL/Pd complex (Fe3O4@MePMO-IL/Pd) was synthesized and characterized, and its catalytic performance was investigated. The preparation of the Fe3O4@MePMO composite was achieved through coating of Fe3O4 nano particles with a mixture of tetramethoxysilane, bis(triethoxysilyl)methane, and (3-chloropropyl)trimethoxysilane in the presence of cetyltrimethylammonium bromide surfactant. The Fe3O4@MePMO was then modified with alkyl imidazolium ionic liquid and palladium species to deliver the Fe3O4@MePMO-IL/Pd nanocatalyst. This catalyst was characterized using Fourier transform infrared, thermal gravimetric, wide-angle powder X-ray diffraction, low-angle powder X-ray diffraction, scanning electron microscopy, transmission electron microscopy, vibrating sample magnetometer, energy-dispersive X-ray, and nitrogen adsorption-desorption analyses. The Fe3O4@MePMO-IL/Pd was effectively used as a highly recoverable and durable catalyst for the selective oxidative coupling of phenols and 2-naphthols under aerobic conditions.
