Chandra, K. Muni;Goud, Nerella Sridhar;Arifuddin, Mohammed;Alvala, Mallika;Alvala, Ravi;Angeli, Andrea;Supuran, Claudiu T. research published 《 Synthesis and biological evaluation of novel 4,7-disubstituted coumarins as selective tumor-associated carbonic anhydrase IX and XII inhibitors》, the research content is summarized as follows. With an aim to develop novel potential anti-cancer agents we have designed, synthesized and evaluated a series of novel 4,7-disubstituted coumarin hybrids for their inhibitory activity against the human carbonic anhydrase isoforms namely CA I, CA II, CA IX and CA XII. The results of CA inhibition clearly showed that the novel 4, 7-disubstituted coumarin hybrids exhibited selective inhibition towards tumor associated isoforms, CA IX and CA XII without inhibiting CA I and CA II isoforms. Among all the compound 8b (I) showed a significant inhibition against hCA IX with a Ki of 0.58μM whereas, the compound 7c (II) showed a significant inhibition against hCA XII with a Ki of 0.36μM resp. All other compounds have shown a good inhibition against hCA IX and hCA XII over hCA I and hCA II within the range of 0.46 to 9.35μM. Therefore, compound 8b and 7c would be the potential leads for developing selective cytotoxic agents targeting hCA IX and XII.

To enhance the biol. activity of the natural product geodin isolated from the marine-derived fungus Aspergillus sp., a series of new ether derivatives I [R = Et, 2-bromobenzyl, 2-chlorobenzyl, etc.] was designed and semisynthesized using a high-yielding one-step reaction. In addition, the insecticidal and antibacterial activities of all geodin congeners I were evaluated systematically. Most of these derivatives I showed better insecticidal activities against Helicoverpa armigera Hubner than Geodin. In particular, I [R = 2,3,4,5-tetrafluorobenzyl] showed potent insecticidal activity with an IC50 value of 89μM, comparable to the pos. control azadirachtin (IC50 = 70μM). Addnl., I [R = 2-chlorobenzyl, 2-fluorobenzyl, 3,5-difluorobenzyl, 2,3,4-trifluorobenzyl, 4-cyanobenzyl and 2-fluoro-3-chlorobenzyl] showed strong antibacterial activity against Staphylococcus aureus and Aeromonas salmonicida with MIC values in the range of 1.15-4.93μM. The preliminary structure-activity relationships indicated that the introduction of halogenated benzyl especially fluorobenzyl, into geodin and substitution of 4-OH was a key factors in increasing the insecticidal and antibacterial activities of geodin.

Cao, Hao;Cheng, Binbin;Liu, Ting;Chen, Jianjun research published 《 Synthesis and pharmacological evaluation of novel resorcinol biphenyl ether analogs as small molecule inhibitors of PD-1/PD-L1 with benign toxicity profiles for cancer treatment》, the research content is summarized as follows. Programmed death protein 1 (PD-1)/programmed death protein ligand 1 (PD-L1) pathway is one of the most actively pursued targets in cancer immunotherapy. In a continuation of authors research interest in this pathway, synthesized and evaluated the pharmacol. activities of a series of resorcinol biphenyl ether analogs as small mol. PD-1/PD-L1 inhibitors for cancer treatment. Among the 27 newly synthesized compounds, I was found to have the highest inhibitory effect against PD-1/PDL-1 with an IC50 value of 56.58 nM in the HTRF (homogeneous time-resolved fluorescence) assay. In addition, I dose-dependently promoted HepG2 cell death in a co-culture model of HepG2/hPD-L1 and Jurkat T cells. Furthermore, mol. modeling study indicated that I binds with high affinity to the binding interface of PD-L1. Moreover, I effectively inhibited tumor growth (TGI of 76.4% at 90 mg/kg) in an immune checkpoint humanized mouse model with no obvious toxicity. Finally, I did not cause in vivo cardiotoxicity and bone marrow suppression (myelosuppression) to BALB/c mice. Taken together, these results suggest that I deserves further investigation as a potent and safe PD-1/PDL-1 inhibitor for cancer treatment.

The bioassay results showed that the target compounds possessed moderate to good fungicidal activities against P. capsici, among which oxime ether compound I shows the highest fungicidal activity in vitro (EC50 = 0.0104μg/mL) which is higher than dimethomorph (EC50 = 0.1148μg/mL) and diacetylenyl amide (EC50 = 0.040μg/mL). Compared with oxime ether compounds (the two nitrogen atoms are on the opposite sides), the activities of oxime ester compounds were significantly reduced. It is different from the com. fungicide fluoxapiprolin, and the activities of the compounds with the two nitrogen atoms on the same side were significantly reduced compared to the compounds with the two nitrogen atoms on the opposite sides. Moreover, some compounds showed moderate to good antifungal activities in vivo against Phytophthora capsici, Pseudoperonospora cubensis, and Phytophthora infestans. SEM of compound I on the hyphae morphol. showed that compound I might cause mycelial abnormalities of P. capsici.

Bai, Li;Xu, Yongqi;Hu, Cheng-Min;Dong, Lei;Wang, Xinping;Li, Wei-Shi;Zhao, Fu-Gang research published 《 Potassium reduced graphite functionalization: Architectural aesthetics and electrical excellence》, the research content is summarized as follows. Rational functionalization plays an important role to push forward graphene applications in multifarious cutting-edge technologies. A key topic that how to program the regular distribution of functional groups, however, remains a big challenge. Herein, we developed a very simple, high-throughput graphite reduction methodol. to attain the neg.-charged graphene which carried ultrahigh-d. and evenly-distributed neg. charges. Guided by these neg. charges, electrophiles were regularly attached to graphene sheets. On the other hand, potassium reduction will not break the carbon-carbon σ-bonds, hence graphene hexagonal lattice was kept as perfect as the pristine pattern. Structural advantages of the neg.-charged graphene derivatives allowed a far more excellent conductivity and electron mobility than the counterparts derived from the prevalent graphene oxide precursor.

Asadizadeh, Saeedeh;Sohrabi, Marzieh;Mereiter, Kurt;Farrokhpour, Hossein;Meghdadi, Soraia;Amirnasr, Mehdi research published 《 Novel octanuclear copper(I) clusters [Cu8{(N)-(μ4-S)}43-I)2I2(PPh3)2] produced via reductive S-S bond cleavage of disulfide Schiff base ligands and their use as efficient heterogeneous catalysts in CuAAC click reaction》, the research content is summarized as follows. Herein, the facile one pot synthesis of novel octanuclear copper(I) complexes, [Cu8{(N)-(μ4-S)}43-I)2I2(PPh3)2], by the reaction of copper(I) iodide and disulfide Schiff base mols. (R-CH=NPhSSPh-N=CH-R, RMePh = 4-MePh, RMeOPh = 4-MeOPh, RMeSPh = 4-MeSPh, RBrPh = 4-BrPh, and RFur = 2-furan) in the presence of PPh3 in dichloromethane-acetonitrile or chloroform-acetonitrile solution under ambient conditions is reported. Single-crystal X-ray structure analyses of these clusters confirm the presence of Cu8S4I4 core stabilized by the chelating Schiff base thiolate fragment R-CH=N-Ph-S produced by the reductive cleavage of disulfide bond, I, and PPh3. DFT calculations have been performed to describe a speculative mechanism of cluster formation. The octanuclear clusters [Cu8{(N)-(μ4-S)}43-I)2I2(PPh3)2] have been exploited as efficient heterogeneous catalysts for the green synthesis of 1,2,3-triazoles from NaN3, alkyl halides, and alkyne derivatives (CuAAC reaction) under mild conditions.
