Fanourakis, Alexander;Williams, Benjamin D.;Paterson, Kieran J.;Phipps, Robert J. research published 《 Enantioselective Intermolecular C-H Amination Directed by a Chiral Cation》, the research content is summarized as follows. A family of anionic variants of the best-in-class catalyst for Rh-catalyzed C-H amination, Rh2(esp)2, with which the chiral cations are associated And derived from quaternized cinchona alkaloids, has been described. These ion-paired catalysts enable high levels of enantioselectivity to be achieved in the benzylic C-H amination of substrates R(CH2)4OH (R = Ph, 1-naphthyl, 3-methylthiophen-2-yl, etc.) bearing pentyl hydroxyl groups. Addnl., the quinoline of the chiral cation appears to engage in axial ligation to the rhodium complex, providing improved yields of products RCH(NHS(O)2OCH2R1)(CH2)3OH (R1 = (CF2)2CF3) vs. Rh2(esp)2 and highlighting the dual role that the cation is playing. These results underline the potential of using chiral cations to control enantioselectivity in challenging transition-metal-catalyzed transformations.

Elgendy, Bahaa;Griffett, Kristine;Hegazy, Lamees;Di Fruscia, Paolo;Sample, Kirby;Schoepke, Emmalie;Kamenecka, Theodore M.;Burris, Thomas P. research published 《 Synthesis and structure activity relationship of the first class of LXR inverse agonists》, the research content is summarized as follows. Liver X Receptors (LXRs) are members of the nuclear receptor family, and they play significant role in lipid and cholesterol metabolism Moreover, they are key regulators of several inflammatory pathways. Pharmacol. modulation of LXRs holds great potential in treatment of metabolic diseases, neurodegenerative diseases, and cancer. We were the first group to identify LXR inverse agonists SR9238 and SR9243 and demonstrate their potential utility in treating liver diseases and cancer. Here, we present the results of structure-activity relationship (SAR) studies, based around SR9238 and SR9243. This study led to identification of I, II, III and IV which were more potent inverse agonists than SR9238 and SR9243 and inhibited expression of the fatty acid synthase gene in DU145 cells. We previously demonstrated that inhibition of FASN is correlated to the anticancer activity of SR9243 and this suggests that new inverse agonists have great potential as anticancer agents. We identified compounds with distinct selectivity toward both LXR isoforms, which can be excellent tools to study the pharmacol. of both isoforms. We employed mol. dynamic (MD) simulations to better understand the mol. mechanism underlying inverse agonist activity and to guide our future design.

Lysine methyltransferases are important regulators of epigenetic signaling and are emerging as a novel drug target for drug discovery. This work demonstrates the positioning of novel 1,5-oxaza spiroquinone scaffold into selective SET and MYND domain-containing proteins 2 methyltransferases inhibitors. Selectivity of the scaffold was identified by epigenetic target screening followed by SAR study for the scaffold. The optimization was performed iteratively by two-step optimization consisting of iterative synthesis and computational studies (docking, metadynamics simulations). Computational binding studies guided the important interactions of the spiro[5.5]undeca scaffold in pocket 1 and Lysine channel and suggested extension of tail length for the improvement of potency (IC50: up to 399 nM). The effective performance of cell proliferation assay for chosen compounds (IC50: up to 11.9 nM) led to further evaluation in xenograft assay. The potent compound 24 demonstrated desirable in vivo efficacy with growth inhibition rate of 77.7% (4 fold decrease of tumor weight and 3 fold decrease of tumor volume). Moreover, mirosomal assay and pharmacokinetic profile suggested further developability of this scaffold through the identification of major metabolites (dealkylation at silyl group, reversible hydration product, the absence of toxic quinone fragments) and enough exposure of the testing compound 24 in plasma. Such spiro[5.5]undeca framework or ring system was neither been reported nor suggested as a modulator of methyltransferases. The chemo-centric target positioning and structural novelty can lead to potential pharmacol. benefit.

Dash, Shruti Gamya;Kantevari, Srinivas;Pandey, Swaroop Kumar;Naik, Pradeep Kumar research published 《 Synergistic interaction of N-3-Br-benzyl-noscapine and docetaxel abrogates oncogenic potential of breast cancer cells》, the research content is summarized as follows. Noscapine, an opium alkaloid, was discovered to bind tubulin, arrest dividing cells at mitosis, and selectively induce apoptosis to cancer cells. N-3-Br-Benzyl-Noscapine (Br-Bn-Nos), one of the derivatives of noscapine, was demonstrated to have improved anticancer potential compared with noscapine. We approached to evaluate the single and combined effect of Br-Bn-Nos and docetaxel (DOX) based on mol. modeling and cellular study. The individual predicted free energy of binding (ΔGbind, pred) for Br-Bn-Nos and DOX with tubulin was found to be -28.89 and -36.07 kcal/mol based on mol. mechanics generalized Born solvation area (MM-GBSA) as well as -26.21 and -34.65 kcal/mol based on mol. mechanics Poisson Boltzmann solvation area (MM-PBSA), resp. However, the ΔGbind, pred of Br-Bn-Nos was significantly reduced (-33.02 and -30.24 kcal/mol using MM-GBSA and MM-PBSA) in the presence of DOX on its binding pocket. Parenthetically, the ΔGbind, pred of DOX was significantly reduced (-37.17 and -32.80 kcal/mol using MM-GBSA and MM-PBSA) in the presence of Br-Bn-Nos on its binding pocket. The reduced ΔGbind, pred in the presence of Br-Bn-Nos and DOX together indicated a combination effect of both the ligands. The combined interaction of both the agents onto tubulin dimmer was also determined exptl. using purified tubulin, in which a combination regimen of Br-Bn-Nos and DOX reduced the fluorescence intensity of tubulin to a higher value (68%) compared with the single regimen. Further, isobologram anal. revealed the synergistic effect of Br-Bn-Nos and DOX in antiproliferative activity using MCF-7 cell line at 48 h (sum FIC = 0.49) and at 72 h (sum FIC = 0.62). The combination dose regimen of Br-Bn-Nos and DOX blocks the cell cycle progression at the G2/M phase and induces apoptosis to cancer cells more effectively compared with the single regimen. Taken together, our study provides compelling evidence that the anticancer potential of noscapine derivatives may be substantially improved when it is used in a combined application with DOX for breast cancer.

Choudhury, Prasun;Chattopadhyay, Shreyasi;De, Goutam;Basu, Basudeb research published 《 Ni-rGO-zeolite nanocomposite: an efficient heterogeneous catalyst for one-pot synthesis of triazoles in water》, the research content is summarized as follows. An important group of pharmaceutical materials, 1,2,3-triazoles, has been synthesized using a Ni-based nanocomposite catalyst (Ni-rGO-zeolite) through azide alkyne cycloaddition (NiAAC). First, a GO-zeolite hybrid was prepared through protonation of a Na-Y-zeolite by H+ ions originating from the -COOH groups of GO. Subsequently the GO-zeolite was treated with Ni-acetate solvothermally in the presence of NaBH4 (reducing atm.). Under the solvothermal conditions a significant part of the incorporated Ni ions in the GO-zeolite were reduced to Ni(0) and simultaneously GO was transformed into rGO. The resulting ternary nanocomposite, Ni-rGO-zeolite, serves as a highly efficient heterogeneous catalyst, and shows excellent regioselectivity forming 1,4-disubstituted-1,2,3-triazoles as the sole product at a low loading of the nickel (∼2.6 mol% with respect to the substrate) with recyclability, and without any significant leaching of the metal. In addition, the Ni-rGO-zeolite exhibits enhanced efficiency under aqueous conditions, proficiency with varying substrates and overcomes some of the shortcomings of the previously reported limited number of Ni-based and other catalysts. The catalytic process is believed to involve the active Ni(0) species, which is stabilized by electron rich rGO that is supported on the microporous high-surface-area zeolite.

Chen, Yan;Huang, Mingzhu;Zhou, Qinghai;Li, Zhen;Meng, Jing;Pan, Mengyuan;Ye, Xiang;Liu, Taifeng;Chang, Shuai;Xiao, Shengxiong research published 《 Regio- and Steric Effects on Single Molecule Conductance of Phenanthrenes》, the research content is summarized as follows. Here, six phenanthrene (the smallest arm-chair graphene nanoribbon) derivatives with dithiomethyl substitutions at different positions as the anchoring groups were synthesized. Scanning tunneling microscopy break junction technique was used to measure their single mol. conductances between gold electrodes, which showed a difference as much as 20-fold in the range of ~10-2.82 G0 to ~10-4.09 G0 following the trend of G2,7 > G3,6 > G2,6 > G1,7 > G1,6 > G1,8. DFT calculations agree well with this measured trend and indicate that the single mol. conductances are a combination of energy alignment, electronic coupling, and quantum effects. This significant regio- and steric effect on the single mol. conductance of phenanthrene model mols. shows the complexity in the practice of graphene nanoribbons as building blocks for future carbon-based electronics in one hand but also provides good conductance tunability on the other hand.

Chen, Yiming;Zhang, Lihui;Zhang, Lin;Jiang, Qixiao;Zhang, Lei research published 《 Discovery of indole-3-butyric acid derivatives as potent histone deacetylase inhibitors》, the research content is summarized as follows. In discovery of HDAC inhibitors (HDACIs) with improved anticancer potency, structural modification was performed on the previous derived indole-3-butyric acid derivative Among all the synthesized compounds, mol. I exhibited high HDAC inhibitory and antiproliferative potencies in the in vitro investigations. The IC50 values of I against HDAC1, HDAC3, and HDAC6 were 13.9, 12.1, and 7.71 nM, resp. In the cancer cell based screening, mol. I showed increased antiproliferative activities in the inhibition of U937, U266, HepG2, A2780, and PNAC-1 cells compared with SAHA. In the HepG2 xenograft model, 50 mg/kg/d of I could inhibit tumor growth in athymic mice compared with 100 mg/kg/d of SAHA. Induction of apoptosis was revealed to play an important role in the anticancer potency of mol.I. Collectively, a HDACI (I) with high anticancer activity was discovered which can be utilized as a lead compound for further HDACI design.
