Eric;Zhang, Haiyan;Jiang, Hualiang;Liu, Hong research published 《 Kinetics-Driven Drug Design Strategy for Next-Generation Acetylcholinesterase Inhibitors to Clinical Candidate》, the research content is summarized as follows. The acetylcholinesterase (AChE) inhibitors remain key therapeutic drugs for the treatment of Alzheimer’s disease (AD). However, the low-safety window limits their maximum therapeutic benefits. Here, a novel kinetics-driven drug design strategy was employed to discover new-generation AChE inhibitors that possess a longer drug-target residence time and exhibit a larger safety window. After detailed investigations, compound 12 was identified as a highly potent, highly selective, orally bioavailable, and brain preferentially distributed AChE inhibitor. Moreover, it significantly ameliorated cognitive impairments in different mouse models with a lower ED than donepezil. The X-ray structure of the cocrystal complex provided a precise binding mode between 12 and AChE. Besides, the data from the phase I trials demonstrated that 12 had good safety, tolerance, and pharmacokinetic profiles at all preset doses in healthy volunteers, providing a solid basis for its further investigation in phase II trials for the treatment of AD.

Zhang, Naichen;Ye, Yuanzhi;Bai, Lu;Liu, Jingjing;Wang, Han;Luan, Xinjun research published 《 Transition metal-free dearomatization of halonaphthols with C(sp3)-electrophiles》, the research content is summarized as follows. The first intermol. electrophilic dearomatization of halonaphthols I (R1 = H, 7-MeO, 6-Me3Si, 3-Cl, etc.) with benzyl/allyl bromides R2Br (R2 = PhCH2, PhCHMe, 4-MeO2CC6H4CH2, H2C:CHCH2, PhCH:CH, etc.) is described. Halonaphthols are used as carbon nucleophiles in dearomatization to form three-dimensional cyclic enones II with excellent chemoselectivity, in which etherification of phenolic hydroxyl group could be restrained by using cesium carbonate as the base. A wide range of cyclic enones was directly prepared from various substituted benzyl/allyl bromides and halonaphthols. Mechanistic investigations suggest a direct SN2 reaction pathway.

The ring-opening difluoromethylation-halogenation of cyclic (thio)ethers is reported through a simple strategy relying on carbon-chalcogen bond activation with difluorocarbene. The reaction proceeds through in situ protonation of the previously little-known difluoromethylene oxonium or sulfonium ylide intermediate followed by ring-opening with halide ion to afford halogenated acyclic difluoromethyl (thio)ethers that can then be employed for further elaboration. TMSCF2X (X=Br, Cl) are unique reagents to achieve this synthetic purpose, which serve as both the difluorocarbene source and the halide ion source.

Zhang, Shuai;Xiao, Jun-Zhao;Li, Yan-Bo;Shi, Chang-Yun;Yin, Liang research published 《 Copper(I)-Catalyzed Asymmetric Alkylation of Unsymmetrical Secondary Phosphines》, the research content is summarized as follows. A Cu(I)-catalyzed asym. alkylation of HPAr1Ar2 with alkyl halides is uncovered, which provides an array of P-stereogenic phosphines in generally high yield and enantioselectivity. The electrophilic alkyl halides enjoy a broad substrate scope, including allyl bromides, propargyl bromide, benzyl bromides, and alkyl iodides. Also, 11 unsym. diarylphosphines (HPAr1Ar2) serve as competent pronucleophiles. The present methodol. is also successfully applied to catalytic asym. double and triple alkylation, and the corresponding products were obtained in moderate diastereo- and excellent enantioselectivities. Some 31P NMR experiments indicate that bulky HPPhMes exhibits weak competitively coordinating ability to the Cu(I)-bisphosphine complex, and thus the presence of stoichiometric HPAr1Ar2 does not affect the enantioselectivity significantly. Therefore, the high enantioselectivity in this reaction is attributed to the high performance of the unique Cu(I)-(R,RP)-TANIAPHOS complex in asym. induction. Finally, one monophosphine and two bisphosphines prepared by the present reaction are employed as efficient chiral ligands to afford three structurally diversified Cu(I) complexes, which demonstrates the synthetic utility of the present methodol.

The major drawbacks of P-glycoprotein (P-gp) inhibitors at the clin. stage make the development of new P-gp inhibitors challenging and desirable. In this study, we reported our structure-activity relationship studies of 4-indolyl quinazoline, which led to the discovery of a highly effective and orally active P-gp inhibitor, YS-370. YS-370 effectively reversed multidrug resistance (MDR) to paclitaxel and colchicine in SW620/AD300 and HEK293T-ABCB1 cells. YS-370 bound directly to P-gp, did not alter expression or subcellular localization of P-gp in SW620/AD300 cells, but increased the intracellular accumulation of paclitaxel. Furthermore, YS-370 stimulated the P-gp ATPase activity and had moderate inhibition against CYP3A4. Significantly, oral administration of YS-370 in combination with paclitaxel achieved much stronger antitumor activity in a xenograft model bearing SW620/Ad300 cells than either drug alone. Taken together, our data demonstrate that YS-370 is a promising P-gp inhibitor capable of overcoming MDR and represents a unique scaffold for the development of new P-gp inhibitors.

Cu2+-crosslinked CM-cellulose/diatomite composite beads (CuII@CMC/DE) were prepared in simple and efficient way using ionic crosslinking techniques. In CuII@CMC/DE beads, diatomite serves as a reinforcing filler, and divalent copper cation acts as crosslinking agent. The chem. structure and morphol. of as-fabricated CuII@CMC/DE beads were well characterized using various techniques such as ICP, FT-IR, XRD, SEM, EDS, elemental mapping, TEM, XPS, and TGA. The CuII@CMC/DE beads exerted superior catalytic performance in CuAAC reactions of benzyl halides, sodium azide, and alkynes to yield regioselective 1,4-disubstituted-1,2,3-triazoles in excellent yields under green conditions. In these resultant catalytic composite beads, Cu2+ was used not only as a crosslinking agent but also as a metal catalyst for CuAAC reaction. The composite beads can be easily separated by filter and reused at least four times without any significant decrease on its activity and selectivity.

Zhou, Yu;Fu, Yan;Yin, Wanchao;Li, Jian;Wang, Wei;Bai, Fang;Xu, Shengtao;Gong, Qi;Peng, Tao;Hong, Yu;Zhang, Dong;Zhang, Dan;Liu, Qiufeng;Xu, Yechun;Xu, H.
