Similarly prepared was 78% 4-bromo-2-(p-nitrobenzal)indan-1,3-dione, m. 283-4° (AcOH). Reaction of 2.27 g. I with 1 g. Ac2CH2, 5 g. Ac2O, and 1 g. Et3N for 24 hrs. gave 0.82 g. III, m. 165-6° (MeOH). Similarly were prepared: 41% IV (with BzCH2Ac), m. 182-3° (EtOH); 53% V (with Bz2CH2), m. 186-7° (EtOAc); 66% VI (with di-Et malonate), m. 126-7° (MeOH). Unlike the condensation product of I with Et acetoacetate, III-VI are stable when refluxed with dilute HCl.

Biol. evaluation showed that these synthesized spiro compounds exhibited moderate to good cytotoxic activity. Napabucasin, undergoing multiple clin. trials, was reported to inhibit the signal transducer and transcription factor 3 (STAT3). To better elucidate its mechanism of action, we designed a napabucasin-based proteolysis targeting chimera (PROTAC), XD2-149(I) that resulted in inhibition of STAT3 signaling in pancreatic cancer cell lines without inducing proteasome-dependent degradation of STAT3. Proteomics anal. of XD2-149 revealed the downregulation of the E3 ubiquitin-protein ligase ZFP91. XD2-149 degrades ZFP91 with DC50 values in the nanomolar range. The cytotoxicity of XD2-149 was significantly, but not fully, reduced with ZFP91 knockdown providing evidence for its multi-targeted mechanism of action. The NQO1 inhibitor, dicoumarol, rescued the cytotoxicity of XD2-149 but not ZFP91 degradation, suggesting that the NQO1-induced cell death is independent of ZFP91. ZFP91 plays a role in tumorigenesis and is involved in multiple oncogenic pathways including NF-κB and HIF-1α.

A series of 20 dual chiral N-cyano sulfilimines containing two centers (carbon and sulfur) were designed and synthesized. All title compounds were determined by 1H NMR, high-resolution mass spectrometry (HRMS) and optical polarimeter. The preliminary results indicated that some of them exhibited favorable insecticidal activities against oriental armyworm (Pseudaletia separata Walker). These isomers exhibited different impact on activity following the sequence as (Sc, Ss) ≥ (Sc, Rs), and the rule of title compounds’ activity against oriental armyworm was 3-CF3 ≥ 2-CH3-4-Cl >2, 3, 4-trifluro in the anilide moiety. The results indicated that these groups such as 3-CF3, 2-CH3-4-Cl or 2, 3, 4-trifluro were inefficient to replace heptafluoroisopropyl group for high larvicidal activity, which provided some guidance for the further modifications of sulfiliminyl dicarboxamides.

Q-switched laser radiation at wavelengths of 355, 532, and 1064 nm from a Nd: YAG laser was used to generate plasma in laboratory air at the target surface made of nano-silver particles of size 95 10 nm. The emitted resonance spectra from the neutral silver at wavelengths of 327.9 nm and 338.2 nm indicate existence of self-reversal in addition to plasma self-absorption. Both lines were identified in emission spectra at different laser irradiation wavelengths with characteristic dips at the un-shifted central wavelengths. These dips are usually associated with self-reversal. Under similar conditions, plasmas at the corresponding bulk silver target were generated. The recorded emission spectra were compared to those obtained from the nano-material target. The comparisons confirm existence of self-reversal of resonance lines that emerge from plasmas produced at nano-material targets. This work suggests a method for recovery of the spectral line shapes and discusses practical examples. In addition, subsidiary calibration efforts that utilize the Balmer series H-line reveal that other Ag I lines at 827.35 nm and 768.7 nm are optically thin under variety of exptl. conditions and are well-suited as reference lines for measurement of the laser plasma electron d.
