New Chalcone Derivative Inhibits ABCB1 in Multidrug Resistant T-cell Lymphoma and Colon Adenocarcinoma Cells

Background/Aim: Development of new potential drugs to overcome multidrug resistance to chemotherapy is a big challenge for cancer treatment. Attention is also given to the natural compounds and their derivatives. The study aimed at evaluating the impact of a new chalcone derivative (1C) on multidrug resistant cell lines, focusing on P-glycoprotein (P-gp, ABCB1) inhibition, as well as 1C-doxorubicin interaction in vitro. Materials and Methods: Cytotoxic and antiproliferative effects of the 1C compound were assessed by thiazolyl blue tetrazolium bromide (MTT) method in mouse T-cell lymphoma and human colon adenocarcinoma cells expressing ABCB1. Alterations in ABCB1 activity were evaluated by rhodamine 123 accumulation assay using flow cytometry. Drug-drug interaction was studied using combination assay. Results: Our results confirmed antiproliferative, cytotoxic, as well as ABCB1 inhibitory potential of 1C in both tested ABCB1-expressing cancer cell lines. Furthermore, 1C displayed synergistic interaction with doxorubicin. Conclusion: Our results suggest the 1C chalcone derivative as a promising compound against resistant lymphoma and colon cancer, which could be used in monotherapy or in combination with other chemotherapeutics.

Assessment of the potential of a reduced dose of dimethyl disulfide plus metham sodium on soilborne pests and cucumber growth

Methyl bromide (MB), a dominant ozone-depleting substance, is scheduled to be completely phased out for soil fumigation by December 30th 2018, in China. The combined effects of dimethyl disulfide (DMDS) plus metham sodium (MNa) were assessed in controlling soilborne pests for soil fumigation. A study was designed in laboratory for the evaluation of the efficacy of DMDS + MNa to control major soilborne pests. At the same time, two trials were conducted in cucumber field located in Tongzhou (in 2012) and Shunyi (in 2013), respectively, in order to assess the potential of DMDS + MNa in controlling soilborne pests. Laboratory studies disclosed positive synergistic effects of almost all four used combinations on Meloidogyne spp., Fusarium spp., Phytophthora spp., Abutilon theophrasti and Digitaria sanguinalis. Field trials found that DMDS + MNa (30 + 21 g a. i. m(-2)), both at a 50% reduced dose, effectively suppressed Meloidogyne spp. with a low root galling index (2.1% and 11.7%), significantly reduced the levels of Phytophthora and Fusarium spp. with a low root disease index (7.5% and 15.8%), gave very high cucumber yields (6.75 kg m(-2) and 10.03 kg m(-2)), and increased income for cucumber growers with the highest economic benefits (20.91 (sic) m(-2) and 23.58 (sic) m(-2)). The combination treatment provided similar results as MB standard dose treatment (40 g a. i. m-2) or DMDS standard dose treatment (60 g a. i. m(-2)) in pest control and yield, but was more effective than MNa standard dose treatment (42 g a. i. m(-2)). Usage of all chemical treatments gave better significant results than the untreated group of control. Considering the economic benefits, the DMDS plus MNa combination (30 + 21 g a. i. m(-2)) could be used for soil fumigation in cucumber production in China.

Simultaneous fabrication of carbon nanodots and hydroxyapatite nanoparticles from fish scale for biomedical applications

Fish industries and markets produce large quantities of fish scales, skins, shells, and bone wastes post processing that contaminate the environment and cause health risks in humans. In this context, we have developed a novel and simple integrated process to valorize the Lethrinus lentjan fish scales by fabricate carbon nanodots (CDs) and hydroxyapatite nanoparticles (HA NPs) simultaneously. The fish scale treatment was carried out by hydrothermal method at 280 degrees C that produced CDs and HA NPs simultaneously. Under hydrothermal treatment, organic and inorganic substances of fish scale is transformed to CDs and HA NPs respectively. As TEM images confirmed that fish scale derived CDs were spherically shaped and similar to 3 to 15 nm in size. The CDs exhibited excitation-dependent emission in photoluminescence. The HA NPs were similar to 8 to 12 nm in diameter and similar to 50 to 100 nm in length with rod shape. Also, HA NPs possess spherical shape nanostructures with 15-50 nm in diameter. Furthermore, we assessed the cytotoxic behavior of synthesized nanostructures using the MTT assay and acridine orange/ethidium bromide (AO/EB) staining. These results showed that synthesized CDs and HA NPs did not cause significant changes in cell viability and morphology, indicating biocompatibility. Additionally, the synthesized CDs and HA NPs were exploited as fluorescent probes for cellular imaging and osteogenic differentiation of stem cells respectively. Overall, the study results indicate that low-cost fish waste was valorized by producing CDs and HA NPs concurrently. The synthesized nanostructures can be applicable for bio-imaging and bone tissue engineering applications.

Various enantiomerically enriched non-proteinogenic (S)-alpha-amino acids were obtained in high chemical and optical yields through Sonogashira cross-coupling reactions and Glaser couplings. The Schiff base Ni(II)complexes of the chiral auxiliary (S)-BPB (BPB = N-benzyl proline benzophenone) and an amino acid containing an aryl bromide moiety in the alpha-position were synthesized and subsequently used in Sonogashira coupling reactions. The obtained Ni(II) complexes were hydrolyzed with aqueous HCl and novel amino acids, containing an alkynylaryl moiety, were isolated in good yields and with high enantiomeric excess (>99% ee). The chiral auxiliary (S)-BPB can be recycled and reused for the synthesis of the starting Ni(II) complex.

Herein, we describe an efficient transition-metal-catalysed process capable of forming P-C bonds from P-4. Using blue-light photocatalysis, this method directly affords valuable triarylphosphines and tetraarylphosphonium salts in a single reaction step.

Enhanced toughness and strength of boron carbide ceramics with reduced graphene oxide fabricated by hot pressing

Boron carbide (B4C) hybrids with different contents of graphene oxide (GO) were prepared by a heterogeneous co-precipitation method using cetyltrimethyl ammonium bromide (CTAB) as the cationic surfactant. The asobtained mixtures were further hot-pressed at 1950 degrees C for 60 min under 30 MPa, by which B4C-reduced GO (rGO) composites were fabricated. It was found that the addition of only 0.5 wt% rGO could alter the predominance of trans-granular fracture in monolithic B4C ceramic material to mixed trans-granular and intergranular modes in B4C-rGO composites. The flexural strength and fracture toughness of the B4C-2 wt% rGO were increased by 31% (from 350 to 455 MPa) and 83% (from 3.20 to 5.85 MPa.m(1/2)), respectively, compared with those of pure B4C. The improved mechanical properties are attributed to the mechanisms of pull-out and bridging of rGO and crack deflection, as evidenced by microstructural observations. The energy dissipation in the present B4C-rGO composites was further verified using two micromechanical models.

Activating ATRP Initiators to Incorporate End-Group Modularity into Photo-RAFT Polymerization

Heterogeneous photocatalysis is increasingly used in reversible deactivation radical polymerization (RDRP). In this study, we found that alkyl bromide redox chemistry typically found in atom transfer radical polymerization (ATRP) can be incorporated in concert with dithiocarbonyl disulfide chemistry into the reversible addition-fragmentation chain transfer (RAFT) process via bismuth oxide photocatalysis. This amalgamation of mechanisms introduces end-group modularity-a new layer of control-into RAFT polymers uniquely enabled by photoredox catalysis. We found that a diversity of functionality can be installed at the alpha-end group via alkyl bromides, while the molecular weight distribution can be tuned seamlessly at the omega-end group through the simultaneous addition of multiple disulfides.
