Wu, Fangrui;Nie, Shenyou;Yao, Yuan;Huo, Tong;Li, Xin;Wu, Xiaowei;Zhao, Jidong;Lin, Yi-Lun;Zhang, Yinjie;Mo, Qianxing;Song, Yongcheng research published 《 Small-molecule inhibitor of AF9/ENL-DOT1L/AF4/AFF4 interactions suppresses malignant gene expression and tumor growth》, the research content is summarized as follows. Chromosome translocations involving mixed lineage leukemia (MLL) gene cause acute leukemia with a poor prognosis. MLL is frequently fused with transcription cofactors AF4 (~35%), AF9 (25%) or its paralog ENL (10%). The AHD domain of AF9/ENL binds to AF4, its paralog AFF4, or histone-H3 lysine-79 (H3K79) methyltransferase DOT1L. Formation of AF9/ENL/AF4/AFF4-containing super elongation complexes (SEC) and the catalytic activity of DOT1L are essential for MLL-rearranged leukemia. Protein-protein interactions (PPI) between AF9/ENL and DOT1L/AF4/AFF4 are therefore a potential drug target. Compound I was identified to be a novel small-mol. inhibitor of the AF9/ENL-DOT1L/ AF4/AFF4 interaction with IC50s of 0.9-3.5μM. Pharmacol. inhibition of the PPIs significantly reduced SEC and DOT1L-mediated H3K79 methylation in the leukemia cells. Gene profiling shows compound I significantly suppressed the gene signatures related to onco-MLL, DOT1L, HoxA9 and Myc. It selectively inhibited proliferation of onco-MLL- or Myc-driven cancer cells and induced cell differentiation and apoptosis. Compound I exhibited strong antitumor activity in a mouse model of MLL-rearranged leukemia. The AF9/ENL-DOT1L/AF4/AFF4 interactions are validated to be an anticancer target and compound I is a useful in vivo probe for biol. studies as well as a pharmacol. lead for further drug development.

Wang, Zongqiang;Forelli, Nicholas;Hernandez, Yozen;Ternei, Melinda;Brady, Sean F. research published 《 Lapcin, a potent dual topoisomerase I/II inhibitor discovered by soil metagenome guided total chemical synthesis》, the research content is summarized as follows. In natural product discovery programs, the power of synthetic chem. is often leveraged for the total synthesis and diversification of characterized metabolites. The synthesis of structures that are bioinformatically predicted to arise from uncharacterized biosynthetic gene clusters (BGCs) provides a means for synthetic chem. to enter this process at an early stage. The recent identification of non-ribosomal peptides (NRPs) containing multiple ρ-aminobenzoic acids (PABAs) led us to search soil metagenomes for BGCs that polymerize PABA. Here, we use PABA-specific adenylation-domain sequences to guide the cloning of the lap BGC directly from soil. This BGC was predicted to encode a unique N-acylated PABA and thiazole containing structure. Chem. synthesis of this structure gave lapcin, a dual topoisomerase I/II inhibitor with nM to pM IC50s against diverse cancer cell lines. The discovery of lapcin highlights the power of coupling metagenomics, bioinformatics and total chem. synthesis to unlock the biosynthetic potential contained in even complex uncharacterized BGCs.

To discover new nematicides I (n = 1, 2, 3; R = Et, iso-Pr, 4-nitrobenzyl, etc.) a series of novel amide derivatives containing an imidazo[1,2-a]pyridine moeity I were designed and synthesized. Among the title compounds, compounds I (n = 1; R = Pr and n = 3; R = n-decyl (III)) exhibited good nematicidal activities against Aphelenchoides besseyi (rice white-tip nematode), with LC50 values against of 27.3 and 35.9 mg/L, resp., which were superior to that of fosthiazate (45.4 mg/L). Meanwhile, the LC50 value of compound III against Caenorhabditis elegans was 5.7 mg/L, which was superior to that of fosthiazate (77.2 mg/L). Compound III not only binds well to acetylcholinesterase (AChE) of nematodes, but also has a good inhibitory activity against AChE. Thus, AChE may be a potential target of compound III against nematodes. Unexpectedly, compound I (n = 1; R = chloromethyl (IV)) exhibited excellent antibacterial activities with EC50 values of 1.2 and 3.1 mg/L against Xanthomonas oryzae pv. oryzae (Xoo) and Xanthomonas oryzae pv. oryzicola (Xoc), resp., which were superior to those of bismerthiazol (68.6 and 77.1 mg/L) and thiodiazole copper (80.8 and 96.6 mg/L). The curative and protective activities of compound IV against bacterial leaf blight were 37.0% and 36.8% at 50 mg/L, resp., which were higher than those of thiodiazole copper (16.1% and 15.5%). In addition, compound IV may inhibit the growth of Xoo by affecting the production of cell membranes and extracellular polysaccharides. Amide derivatives containing an imidazo[1,2-a]pyridine moeity Ican be used as good lead-structures to discover new nematicidal and antibacterial agents in the future.

Wang, Haibin;Xu, Xin;Yin, Jichen;Zhang, Zhifeng;Xue, Lei research published 《 A Highly Selective “Turn-On” Fluorescent Sensor for Aluminum Ion Detection in Aqueous Solution Based on Imidazo[2,1-b]thiazole Schiff Base》, the research content is summarized as follows. Low sensitivity of fluorescent sensors with “turn-off” response for identifying Al3+ has limited their wide application in the field of ion sensing. Herein, this paper reports a novel fluorescence sensor (L) endowing with “turn-on” fluorescence response based on imidazo[2,1-b]thiazole for recognizing Al3+ in aqueous solutions Results show that the L exhibits high selectivity and sensitivity with “turn-on” fluorescence response towards Al3+, but also has strong anti-interference to other common ions. In addition, the excellent color change can be distinguished by naked eyes under UV light and the test papers show great potential for detecting Al3+ in tap water. The supposed sensing mechanism of L to Al3+ could be attributed to the inhibition of photo-induced electron transfer (PET) and chelation-enhanced fluorescence (CHEF). Such a novel design pathway is expected to offer helpful guidance for fabricating various sensors with “turn-on”; fluorescence response.

Herein, we describe a simple and robust cis-selective hydrogenation of abundant and cheap fluoropyridines to yield a broad scope of (multi)fluorinated piperidines. This protocol enables the chemoselective reduction of fluoropyridines while tolerating other (hetero)aromatic systems using a com. available heterogeneous catalyst. Fluorinated derivatives of important drug compounds are prepared, and a straightforward strategy for the synthesis of enantioenriched fluorinated piperidines is disclosed.

Wang, Ao;Wang, Yawan;Meng, Xin;Yang, Yushe research published 《 Design, synthesis and biological evaluation of novel thiohydantoin derivatives as potent androgen receptor antagonists for the treatment of prostate cancer》, the research content is summarized as follows. Two series of novel thiohydantoin derivatives of enzalutamide I (X = C, N, S; R = Br, Ph, 3-pyridyl) and II (M = C, N; A = di-Me, cyclobutyl, oxetanyl, etc.; X = C, N; Y = O, N-methyl; n = 0, 1) as potent androgen receptor (AR) antagonists were designed and synthesized. Among them, compound II (M = C; A = dimethyl; X = C; Y = O; n =1) was identified as an AR antagonist which was 2.3-fold more potent than enzalutamide. Mol. docking studies were performed to explain the improved potency of II (M = C; A = dimethyl; X = C; Y = O; n =1) at AR. In cell proliferation assays, II (M = C; A = dimethyl; X = C; Y = O; n =1) exhibited similar anti-proliferative activities with enzalutamide against hormone sensitive LNCaP cells and AR-overexpressing LNCaP/AR cells. These data indicated that II (M = C; A = dimethyl; X = C; Y = O; n =1) can be a good lead compound for further structure optimization for the treatment of prostate cancer.

Thiazole has been less focused, compared to thiophene, as the building unit for π-conjugated polymers that are used in organic electronic devices despite its electron deficiency and ability to coplanarize the polymer backbone, which are crucial for design of such π-conjugated polymers. Here, we show that bithiazole dicarboxylate ester (BETz), which is quite easily synthesized, is a promising building unit for π-conjugated polymers. Copolymers based on BETz, having unsubstituted bithiophene and bithiazole as counits, indeed form highly crystalline structures in thin films, which is beneficial for semiconducting properties. Of particular interest is that the BETz-based polymers are found to show electron-transport property even though the LUMO energy levels are not very low, as they show ambipolar and unipolar n-channel behaviors depending on the counit in organic transistor devices. We believe that BETz allows us to design a wide variety of π-conjugated polymers that provide fascinating functions in organic electronic devices.

In this research, a novel procedure for the synthesis of pyrazole derivatives in excellent yields was studied using catalyst-free multicomponent reaction of isoquinoline, activated acetylenic compounds, alkyl bromides, triphenylphosphine and hydrazine in water under ultrasonic irradiation at room temperature The advantages of this procedure than to reported methods are short time of reaction, high yields of product, easy separation of product, clean mixture of reaction and green media for performing reaction. In addition, because of having pyrazole and isoquinoline core in the synthesized compounds, in this research, antioxidant activity of some synthesized compounds was investigated.

70-23-5, Ethyl bromopyruvate molecular formula is C5H7BrO3 and its molecular weight is 195.01 g/mol. The purity is usually 95%.

Ethyl bromopyruvate is used in a synthesis of thioxothiazolidines from carbon disulfide and primary amines.

Ethyl bromopyruvate is a chemical inhibitor that inhibits the enzyme pyruvate dehydrogenase, which is responsible for the conversion of pyruvic acid to acetyl-CoA. This inhibition leads to a decrease in ATP levels and can cause metabolic disorders. Ethyl bromopyruvate is used as an anthelmintic drug and in asymmetric synthesis. It is also used in the synthesis of thiostrepton, an antibiotic that has been shown to have antimicrobial activity against Gram-positive bacteria, including Staphylococcus aureus and Streptococcus pneumoniae., Computed Properties of 70-23-5
