A series of phenylthiazole and phenylthiophene pyrimidindiamine derivatives l [R1 = ethylamino, propylamino, butylamino, pyrrolidino; R2 = H, OMe, F, Cl, Br, etc.] were designed and synthesized by modifying the hit compound (N2-isobutyl-N4-((4-methyl-2-phenylthiazol-5-yl)methyl) pyrimidine-2,4-diamine) and their antibacterial activities were evaluated both in-vitro and in-vivo was reported. Among the tested compounds, compound I [R1 = isobutylamino; R2 = 3-bromo] displayed the best antibacterial activities, which was not only capable of inhibiting E. coli and S. aureus growth at concentrations as low as 2 and 3μg/mL in-vitro, but also efficacious in a mice model of bacteremia in vivo. Unlike conventional antibiotics, compound I [R1 = isobutylamino; R2 = 3-bromo] was elucidated to mainly destroy the bacterial cell membrane, with the dissipation of membrane potential and leakage of contents, ultimately leading to cell death. The destruction of cell structure was challenging to induce bacterial resistance, which suggested that compound I [R1 = isobutylamino; R2 = 3-bromo] was a promising alternatives to antibiotics against bacteria.

Bromodomain and extra-terminal (BET) is a promising therapeutic target for various hematol. cancers. We used the BRD4 inhibitor compound 13 (I) as a lead compound to develop a variety of compounds, and we introduced diverse groups into the position of the compound 13 orienting toward the ZA channel. A series of compounds bearing triazolopyridazine motif exhibited remarkable BRD4 protein inhibitory activities. Among them, compound 39 (II) inhibited BRD4(BD1) protein with an IC50 of 0.003μM which was superior to lead compound 13. Meanwhile, compound 39 possess activity, IC50 = 2.1μM, in antiproliferation activity against U266 cancer cells. On the other hand, compound 39 could arrest tumor cells into the G0/G1 phase and induce apoptosis, which was consistent with its results in inhibiting cell proliferation. Biol. and biochem. data suggest that BRD4 protein might be a therapeutic target and that compound 39 is an excellent lead compound for further development.

Mitochondria are pivotal energy production sources for cells to maintain necessary metabolism activities. Targeting dysfunctional mitochondrial features has been a hotspot for mitochondrial-related disease researches. Investigation with cancerous mitochondrial metabolism is a continuing concern within tumor therapy. Herein, we set out to assess the anti-cancer activities of a novel family of TPP-thiazole derivatives based on our earlier research on mitochondrial targeting agents. Specifically, we designed and synthesized a series of TPP-thiazole derivatives and revealed by the MTT assay that most synthesized compounds effectively inhibited three cancer cell lines (HeLa, PC3 and MCF-7). After structure modifications, we explored the SAR relationships and identified the most promising compound R13 (IC50 of 5.52μM) for further investigation. In the meantime, we performed ATP production assay to assess the selected compounds inhibitory effect on HeLa cells energy production The results displayed the test compounds significantly restrained ATP production of cancer cells. Overall, we have designed and synthesized a series of compounds which exhibited significant cytotoxicity against cancer cells and effectively inhibited mitochondrial energy production

Acute lung injury/acute respiratory distress syndrome (ALI/ARDS) is one of the most common complications in COVID-19. Elastase has been recognized as an important target to prevent ALI/ARDS in the patient of COVID-19. Cyclotheonellazole A (CTL-A) is a natural macrocyclic peptide reported to be a potent elastase inhibitor. Herein, we completed the first total synthesis of CTL-A in 24 linear steps. The key reactions include three-component MAC reactions and two late-stage oxidations We also provided seven CTL-A analogs and elucidated preliminary structure-activity relationships. The in vivo ALI mouse model further suggested that CTL-A alleviated acute lung injury with reductions in lung edema and pathol. deterioration, which is better than sivelestat, one approved elastase inhibitor. The activity of CTL-A against elastase, along with its cellular safety and well-established synthetic route, warrants further investigation of CTL-A as a candidate against COVID-19 pathogeneses.

Damghani, Tahereh;Moosavi, Fatemeh;Khoshneviszadeh, Mehdi;Mortazavi, Motahareh;Pirhadi, Somayeh;Kayani, Zahra;Saso, Luciano;Edraki, Najmeh;Firuzi, Omidreza research published 《 Imidazopyridine hydrazone derivatives exert antiproliferative effect on lung and pancreatic cancer cells and potentially inhibit receptor tyrosine kinases including c-Met》, the research content is summarized as follows. Abstract: Aberrant activation of c-Met signalling plays a prominent role in cancer development and progression. A series of 12 imidazo [1,2-α] pyridine derivatives bearing 1,2,3-triazole moiety were designed, synthesized and evaluated for c-Met inhibitory potential and anticancer effect. The inhibitory activity of all synthesized compounds against c-Met kinase was evaluated by a homogeneous time-resolved fluorescence (HTRF) assay at the concentration range of 5-25 μM. Derivatives 6d, 6e and 6f bearing Me, tert-Bu and dichloro-Ph moieties on the triazole ring, resp., were the compounds with the highest potential. They significantly inhibited c-Met by 55.3, 53.0 and 51.3%, resp., at the concentration of 25 μM. Synthetic compounds showed antiproliferative effects against lung (EBC-1) and pancreatic cancer cells (AsPc-1, Suit-2 and Mia-PaCa-2) expressing different levels of c-Met, with IC50 values as low as 3.0 μM measured by sulforhodamine B assay. Active derivatives significantly blocked c-Met phosphorylation, inhibited cell growth in three-dimensional spheroid cultures and also induced apoptosis as revealed by Annexin V/propidium iodide flow cytometric assay in AsPc-1 cells. They also inhibited PDGFRA and FLT3 at 25 μM among a panel of 16 kinases. Mol. docking and dynamics simulation studies corroborated the exptl. findings and revealed possible binding modes of the select derivatives with target receptor tyrosine kinases. The results of this study show that some imidazopyridine derivatives bearing 1,2,3-triazole moiety could be promising molecularly targeted anticancer agents against lung and pancreatic cancers.

Bur, Scott K.;Pomerantz, William C. K.;Bade, Morgan L.;Gee, Clifford T. research published 《 Fragment-Based Ligand Discovery Using Protein-Observed 19F NMR: A Second Semester Organic Chemistry CURE Project》, the research content is summarized as follows. Curriculum-based undergraduate research experiences (CUREs) have been shown to increase student retention in STEM fields and are starting to become more widely adopted in chem. curricula. Here we describe a 10-wk CURE that is suitable for a second-semester organic chem. laboratory course. Students synthesize small mols. and use protein-observed 19F (PrOF) NMR to assess the small mol.’s binding affinity to a target protein. The research project introduced students to multistep organic synthesis, structure-activity relationship studies, quant. biophys. measurements (measuring Kd from PrOF NMR experiments), and scientific literacy. Docking experiments could be added to help students understand how changes in a ligand structure may affect binding to a protein. Assessment using the CURE survey indicates self-perceived skill gains from the course that exceed gains measured in a traditional and an inquiry-based laboratory experience. Given the speed of the binding experiment and the alignment of the synthetic methods with a second-semester organic chem. laboratory course, a PrOF NMR fragment-based ligand discovery lab can be readily implemented in the undergraduate chem. curriculum.

Ketol-acid reductoisomerase (KARI), the second enzyme in the branched-chain amino acid biosynthesis pathway, is a potential drug target for bacterial infections including Mycobacterium tuberculosis. Here, we have screened the Medicines for Malaria Venture Pathogen Box against purified M. tuberculosis (Mt) KARI and identified two compounds that have Ki values below 200 nM. In Mt cell susceptibility assays one of these compounds exhibited an IC50 value of 0.8μm. Co-crystallization of this compound, 3-((methylsulfonyl)methyl)-2H-benzo[b][1,4]oxazin-2-one (MMV553002), in complex with Staphylococcus aureus KARI, which has 56% identity with Mt KARI, NADPH and Mg2+ yielded a structure to 1.72 Å resolution However, only a hydrolyzed product of the inhibitor (i.e. 3-(methylsulfonyl)-2-oxopropanic acid, missing the 2-aminophenol attachment) is observed in the active site. Surprisingly, Mt cell susceptibility assays showed that the 2-aminophenol product is largely responsible for the anti-TB activity of the parent compound Thus, 3-(methylsulfonyl)-2-oxopropanic acid was identified as a potent KARI inhibitor that could be further explored as a potential biocidal agent and we have shown 2-aminophenol, as an anti-TB drug lead, especially given it has low toxicity against human cells. The study highlights that careful anal. of broad screening assays is required to correctly interpret cell-based activity data.

It is also used in the synthesis of thiostrepton, an antibiotic that has been shown to have antimicrobial activity against Gram-positive bacteria, including Staphylococcus aureus and Streptococcus pneumoniae., 70-23-5.

Vinyl bromides undergo the Heck reaction, which involves C-C coupling with alkene to give substituted alkenes. 70-23-5, formula is C5H7BrO3, Name is Ethyl 3-bromo-2-oxopropanoate. Methyl bromide is a precursor in the manufacture of several chemicals and is employed as a soil sterilant, mainly for seed production. Formula: C5H7BrO3.

Babaev, Eugene V.;Nevskaya, Aleksandra A.;Dlynnikh, Ilya V.;Rybakov, Victor B. research published 《 Synthesis of antiaromatic thiazinoindolizines based on electrophilic cyclizations of indolizine-5-thione》, the research content is summarized as follows. Alkylation of indolizinethione I (Ar = 4-methylphenyl) at the sulfur atom by the action of RCOCH2Br (R = Me, ethoxycarbonyl, 4-chlorophenyl, etc.) gave thioethers which on subsequent ring closure and dehydration lead to the formation of stable antiarom. thiazino[4,3,2-cd]indolizines II.

Formula: C5H7BrO3, Ethyl bromopyruvate molecular formula is C5H7BrO3 and its molecular weight is 195.01 g/mol. The purity is usually 95%.

Ethyl bromopyruvate is used in a synthesis of thioxothiazolidines from carbon disulfide and primary amines.
