Structural characterization and in vitro lipid binding studies of non-specific lipid transfer protein 1 (nsLTP1) from fennel (Foeniculum vulgare) seeds

Non-specific lipid transfer proteins (nsLTPs) are cationic proteins involved in intracellular lipid shuttling in growth and reproduction, as well as in defense against pathogenic microbes. Even though the primary and spatial structures of some nsLTPs from different plants indicate their similar features, they exhibit distinct lipid-binding specificities signifying their various biological roles that dictate further structural study. The present study determined the complete amino acid sequence, in silico 3D structure modeling, and the antiproliferative activity of nsLTP1 from fennel (Foeniculum vulgare) seeds. Fennel is a member of the family Umbelliferae (Apiaceae) native to southern Europe and the Mediterranean region. It is used as a spice medicine and fresh vegetable. Fennel nsLTP1 was purified using the combination of gel filtration and reverse-phase high-performance liquid chromatography (RP-HPLC). Its homogeneity was determined by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and matrix-assisted laser desorption/ionization-time of flight (MALDI-TOF) mass spectrometry. The purified nsLTP1 was treated with 4-vinyl pyridine, and the modified protein was then digested with trypsin. The complete amino acid sequence of nsLTP1 established by intact protein sequence up to 28 residues, overlapping tryptic peptides, and cyanogen bromide (CNBr) peptides. Hence, it is confirmed that fennel nsLTP1 is a 9433 Da single polypeptide chain consisting of 91 amino acids with eight conserved cysteines. Moreover, the 3D structure is predicted to have four alpha -helices interlinked by three loops and a long C-terminal tail. The lipid-binding property of fennel nsLTP1 is examined in vitro using fluorescent 2-p-toluidinonaphthalene-6-sulfonate (TNS) and validated using a molecular docking study with AutoDock Vina. Both of the binding studies confirmed the order of binding efficiency among the four studied fatty acids linoleic acid>linolenic acid>Stearic acid>Palmitic acid. A preliminary screening of fennel nsLTP1 suppressed the growth of MCF-7 human breast cancer cells in a dose-dependent manner with an IC50 value of 6.98 mu M after 48 h treatment.

Hot water extract of Glehnia littoralis leaf showed skin-whitening and anti-wrinkle properties

Ethnopharmacological relevance: Glehnia littoralis (Umbelliferae) is a commonly used traditional medicine to treat the immune-related diseases in Korea, China, and Japan. Aim of the study: This study was designed to investigate the elastase and tyrosinase inhibitory potentials of hot water extracts of G. littoralis leaves. Materials and methods: An MTT (3-(4,5-dimethylthiazol-2-yl)-2,5 diphenyltetrazolium bromide) assay was conducted to measure the cytotoxicity of the extract on the HS68 and B16F10 cell lines. An ultraviolet (UV) irradiation protection effect on HS68 cells of the extract was also examined. The elastase and tyrosinase inhibition assays were conducted to determine anti-wrinkle and skin-whitening potentials of the plant extract. Results: The hot water extracts of G. littoralis leaves were nontoxic to two cell lines, HS68 and B16F10. Less than 10% cell death of the two cell lines and remarkable effect of the leaf extracts on UV irradiation protection of HS68 cells indicated noncytotoxic properties of G. littoralis leaves. The leaf extracts were found to have considerable tyrosinase and elastase inhibition potential, demonstrating skin whitening and anti-wrinkle properties, relative to ascorbic (250 mu g/mL) and ursolic (250 mu g/mL) acids. Conclusions: Findings of this study revealed that G. littoralis leaves could be used in the preparation of cosmetic products for skin whitening and anti-wrinkle applications. (C) 2019 SAAB. Gold nanorods-trypsin biocorona: a novel nano composite for in vitro cytotoxic activity towards MCF-7 and A-549 cancer cells

In the present work, we have synthesized cetyltrimethyl ammonium bromide (CTAB) capped gold nanorods (Au NRs) to evaluate apparent binding affinities for the adsorption of trypsin (TRP). The interaction between TRP and Au NRs is explained elaborately using high resolution transmission electron microscopy (HR-TEM), dynamic light scattering (DLS), surface charge distributions and EDAX spectroscopy. The results from the emission spectral measurements explored the existence of strong binding between TRP and Au NRs and confirmed the complex formation via a static quenching process. Moreover, the cytotoxicity of Au NRs and the TRP-Au NR complex on human breast cancer (MCF-7) and lung cancer (A-549) was examined by the [3-((4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazoliumbromide)] MTT assay. Microscopic analysis of nuclear fragmentation studies validates the significant reduction of viability of TRP-Au NRs treated MCF-7 cells and A-549 cells through apoptotic-mediated pathways. These studies demonstrated that the TRP-Au NRs system displays an advanced level of cytotoxicity in cancer cells compared to Au NR-treated MCF-7 cells and A-549 cells.

The role of S100B and nitric oxide in the apoptotic action of pentylenetetrazole on astrocytes

Introduction: Astrocyte, S100B and nitric oxide may have a role in the pathogenesis and treatment of epilepsy. However, the effects of nitric oxide and S100B on the gliotoxic effects of chemical convulsants such as pentylenetetrazole (PTZ) is unknown. Therefore, we aimed to evaluate the effects of S100B and nitric oxide on gliotoxicity of PTZ in a 1321NI1 astrocytic culture. Methods: The 1321N1 astrocytes were exposed to PTZ (40mM), arundic acid (50 mu M) or both of them for 24h. In addition, we poured L-arginine (100 or 500 mu M), N-nitro-L-arginine methyl ester (100 or 500 mu M), 7-nitroindazole (30 or 100 mu M) and aminoguanidine (50 or 100 mu M) to the culture media contained PTZ, arundic acid or both of them and incubated for 24h. Cell viability was measured by the methylthiazolyldiphenyl-tetrazolium bromide reagent and the S100B protein level was measured using an enzyme-linked immunosorbent assay. Results: There was a negative correlation between cell viability in astrocytes and the intracellular S100B levels. PTZ decreased cell viability, but it increased the intracellular S100B levels. Arundic acid, N-nitroarginine methyl ester, 7-nitroindazole and aminoguanidine reversed the PTZ effects on cell viability and intracellular S100B levels. Adding the L-arginine to PTZ plus arundic acid reduced the modulatory effects of arundic acid on PTZ. Conclusion: Nitric oxide and S100B have a role in gliotoxicity of PTZ in cell culture. Arundic acid suppresses PTZ-induced S100B elevation and gliotoxicity possibly by modulation of the nitric oxide pathway.

Biofilm-Forming Ability of Pathogenic Bacteria Isolated from Retail Food in Poland

Biofilms have a significant impact on food safety in the food industry. Many foodborne outbreaks have been associated with pathogenic bacterial strains that can form a biofilm. The present study was conducted under the Official Control and Monitoring Program in Poland to examine the ability of pathogenic bacteria collected from retail food samples to form biofilms. Biofilm formation was assessed by qualitative detection of extracellular polymeric substances on Congo red agar, by adherence to glass with the tube method, by the crystal violet biofilm (CV) assay, and by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay. A total of 40 isolates from food samples (10 strains each of Listeria monocytogenes, Staphylococcus aureus, Escherichia coli, and Bacillus cereus) were examined. The strains were classified as adherent, slightly adherent, or nonadherent; biofilm production was classified as weak (WBP), moderate (MBP), or strong (SBP); and metabolic activity was classified as weak (WMA), moderate (MMA), or high (HMA). The incubation conditions and time influenced the amount of biofilm formed as well as did the growth medium. In the test tubes with Luria-Bertani broth (LBB), 22.5% of the strains were adherent and 77.5% were slightly adherent. Stronger adhesion was obtained in brain heart infusion (BHI) with 2% sucrose; 60% of the isolates were classified as adherent. With the CV assay with LBB, SBP was noted for 7.5% of the strains after 24 h of incubation and for 37.5% of the strains after 48 h. In BHI plus 2% sucrose, SBP was noted for 42.5 and 37.6% of the strains after 24 and 48 h, respectively. With the MTT assay with LBB, HMA was found for 15% of the strains after 24 h of incubation and for 25% of the strains after 48 h. In BHI plus 2% sucrose, 70 and 85% of the incubated strains were classified as HMA after 24 and 48 h, respectively.

Catalytic activity of Pd dithiolate complexes with large-bite-angle diphosphines in Heck coupling reactions

Palladium(II) complexes of aryl dithiolates and wide-bite-angle diphosphines Xantphos and dppf have been developed as efficient catalysts in Suzuki and Suzuki carbonylation reactions. The catalytic activity of these highly stable, discrete and charged complexes was investigated in Heck coupling reactions of styrene and a variety of aryl bromides. Under optimized reaction conditions these palladium complexes showed excellent activity with high turnover number (6 x 10(6)) and high turnover frequency (4 x 10(5) h(-1)). The effect of bite angle of diphosphines on the catalytic activity of the complexes [Pd-2((PP)-P-boolean AND)(2)(SC12H8S)](2)(OTf)(4) followed the trend (PP)-P-boolean AND = Xantphos > dppf > dppe as the order of their bite angles. The catalyst could be reused, and after three cycles the formation of significant amount of Pd nanoparticles was noticed, which were characterized using powder X-ray diffraction, energy-dispersive X-ray analysis and transmission electron microscopy. The high catalytic activity has been attributed to the Pd nanoparticles.

Rare concurrent ocular myasthenia gravis and Graves’ ophthalmopathy in a man with Poland syndrome: a case report

Background Ocular myasthenia gravis and Graves’ ophthalmopathy are autoimmune diseases that are mediated by membrane receptors and share many identical clinical processes. Poland syndrome is a rare congenital deformity characterized by defects of the ipsilateral hand and the chest wall, and it is usually associated with hypoplasia of ipsilateral pectoral muscles and homolateral breast. However, to the best of our knowledge, the co-occurrence of these diseases has never been reported. In this study, we present a man with Poland syndrome who was diagnosed with Graves’ ophthalmopathy and ocular myasthenia gravis in succession. Case presentation A 43-year-old man presented with bilateral upper eyelid ptosis, bilateral eye protrusion, bilateral eye movement disorder and malformation of the right hand. Asymmetrical malformation of the chest wall and ipsilateral hand deformity were shown as Poland syndrome. He was diagnosed with ocular myasthenia gravis and Graves’ ophthalmopathy on the basis of clinical manifestations and laboratory examinations, including bilateral exophthalmos and progressive asymmetrical ophthalmoparesis without pupillary dysfunction, positive autoantibody tests, repetitive nerve stimulation tests, and computed tomography scans. Treatments with pyridostigmine bromide, thymectomy, and prednisone led to partial clinical improvement. After 13 months of follow-up, the symptoms of drooping eyelids were partially improved, but the eyeball protrusion and right hand deformity remained unchanged. Conclusions We report the first case of co-occurrence of ocular myasthenia gravis, Graves’ ophthalmopathy, and Poland syndrome. Genetic predisposition and immune dysregulation might be the pathogenesis of the association.

Understanding the Basics of Electron Transfer and Cyclic Voltammetry of Potassium Ferricyanide – An Outer Sphere Heterogeneous Electrode Reaction

Understanding and controlling the processes occurring at electrode/electrolyte interface are very important in optimizing energy conversion devices. Cyclic voltammetry is a very sophisticated and accurate electroanalytical method enables us to explore the mechanism of such electrode reactions. In this work, electrochemical experiments using cyclic voltammetry were performed in aqueous KCl solution containing potassium ferricyanide, K-3[Fe(CN)(6)] at a glassy carbon disc working electrode and the mechanism of the reactions is highlighted. The CV measurements shows that the ferricyanide [Fe(CN)(6)](3-) reduction to ferrocyanide [Fe(CN)(6)](4-) and the reverse oxidation process follows an outer sphere electrode reaction mechanism. Voltammetry analysis further indicates that the reaction is reversible and diffusion controlled one electron transfer electrochemical process. The peak currents due to [Fe(CN)(6)](4-) oxidation and the peak current due to [Fe(CN)(6)](3-) reduction increase with increase of concentration and scan rate increase. The diffusion co-efficient was determined by applying Randles-Sevcik equation. The report could be helpful for university students to understand the basics of electron transfer in redox processes and cyclic voltammetry.
