Self-Assembled Surfactant-Templated Synthesis of Porous Hollow Silica Nanoparticles: Mechanism of Formation and Feasibility of Post-Synthesis Nanoencapsulation

SiO2 is bioinert and highly functionalizable, thus making it a very attractive material for nanotechnology applications such as drug delivery and nanoencapsulation of pesticides. Herein, we synthesized porous hollow SiO2 nanoparticles (PHSNs) by using cetyltrimethylammonium bromide (CTAB) and Pluronic P123 as the structure-directing agents. The porosity and hollowness of the SiO2 structure allow for the protective and high-density loading of molecules of interest inside the nanoshell. We demonstrate here that loading can be achieved post-synthesis through the pores of the PHSNs. The PHSNs are monodisperse with a mean diameter of 258 nm and a specific surface area of 287 m(2) g(-1). The mechanism of formation of the PHSNs was investigated using 1-D and 2-D solid-state nuclear magnetic resonance (SS-NMR) and Fourier-transform infrared spectroscopy (FTIR). The data suggest that CTAB and Pluronic P123 interact, forming a hydrophobic spherical hollow cage that serves as a template for the porous hollow structure. After synthesis, the surfactants were removed by calcination at 550 degrees C and the PHSNs were added to an Fe3+ solution followed by addition of the reductant NaBH4 to the suspension, which led to the formation of Fe(0) NPs both on the PHSNs and inside the hollow shell, as confirmed by transmission electron microscopy imaging. The imaging of the formation of Fe(0) NPs inside the hollow shell provides direct evidence of transport of solute molecules across the shell and their reactions within the PHSNs, making it a versatile nanocarrier and nanoreactor.

Background: The dysfunction of vascular smooth muscle cells (VSMCs) contributes to the development of atherosclerosis. This study aimed to investigate the role of circular RNA-0010283 (circ_0010283) in oxidized low-density lipoprotein (ox-LDL)-treated VSMCs and the associated action mechanism. Methods and Results: The expression of circ_0010283 was investigated using quantitative real-time polymerase chain reaction (qRT-PCR). Cell proliferation was monitored by using a 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay. Cell apoptosis was detected by using flow cytometry assay. A transwell assay was performed to observe migration and invasion, and a scratch assay was implemented to test migration. The expression of proliferation, apoptosis and migration/invasion-related proteins was measured by using a western blot. The targeted relationship was predicted by using a bioinformatics tool (Starbase) and verified by using a dual-luciferase reporter assay, a RNA immunoprecipitation (RIP) assay and a RNA pull-down assay. circ_0010283 was highly expressed in serum samples from atherosclerosis patients and ox-LDL-treated human VSMCs (HVSMCs). circ_0010283 knockdown suppressed ox-LDL-induced proliferation, migration and invasion in HVSMCs. Effect of prescribed fires on the export of dissolved organic matter, precursors of disinfection by-products, and water treatability

In this study, we report for the first time the effect of prescribed fires on the export of dissolved organic matter (DOM) and precursors of disinfectant by-products (DBPs) from periodically (every 2-3 years) and seasonally (i.e., dormant and growing) burned forest fuel materials (i.e., live vegetation, woody debris, and detritus [litter and duff]) and treatability of its rainwater leachate. Periodically applied (every 2-3 years for 40 years) prescribed fires decreased total fuel load (62 10%), primarily detrital mass (75 +/- 2%). How-ever, functional groups (i.e., phenolic compounds, proteins, carbohydrates, aromatic [1-ring], polycyclic aromatic hydrocarbons [PAHs], and lipids) attached to DOM of ground solid materials did not change significantly. Outside rainwater leaching (from forest fuel materials) experiments showed that the leaching capacity of dissolved organic carbon (DOC) from burned litter samples decreased by 40 +/- 20% regardless of burning season when compared to unburned litter samples. The leaching of total dissolved nitrogen (TDN), dissolved organic nitrogen (DON), ammonium (NH4+), and reactive phosphorus (PO43-) from burned materials decreased between 40 and 70% when compared to unburned materials. Also, DOM composition was affected by prescribed fire, which partially consumed humic-like substances based on fluorescence analyses. Thus, periodically applied prescribed fires also resulted in a reduction of trihalomethane (THM) (42 +/- 23%) and haloacetic acid (HAA) (42 +/- 20%) formation potentials (FPs), while DOC normalized reactivity of THM and HAA FPs did not change significantly. Additionally, the leaching of Nnitrosodimethylamine (NDMA) precursors, bromide ion (Br-), and selected elements (K, Ca, Mg, Mn, Fe, S, Na, B, and Al) were not significantly affected by prescribed fires. Finally, coagulant (i.e., alum and ferric) dose requirements and coagulation efficiencies were similar (i.e., removal of DOC, precursors of THMs and HAAs were 52-56%, 69-70%, 78-79%, respectively) in unburned and pre-burned leachate samples. Leishmaniasis is a notorious disease affecting mainly poor people. Visceral Leishmaniasis (VL) is a clinical form of Leishmaniasis which mainly affects people living below poverty line. There are no available vaccines against VL and chemotherapy still remains the mainstay for treatment. However, the use of the current therapeutic agents are limited due to issues of toxicity, side effects and cost effectiveness. Thus, repositioning of FDA approved drugs for anti-leishmanial therapeutics and encapsulation of the same in nanoliposomes may pave the way of future anti-leishmanial therapeutics. In this work, liposome encapsulated Halofantrine (Halolipo) was prepared by extrusion of hydrated lipid film method. The Halolipo was characterized by Dynamic Light Scattering and Transmission Electron Microscopy. The effect of Halolipo on Leishmania donovani Ag83 promastigotes was assessed by MIT assay and qualitative analysis of ROS generation and mitochondrial membrane depolarization was performed. The cell death was visualized by dual staining with Acridine orange and Ethidium bromide followed by fluorescence microscopy. The encapsulation of Halofantrine drug in liposome was confirmed by characteristic peak at 260 nm by UV-vis spectrophotometer. The formulated halolipo was 20 nm in size and stable for > 4 weeks at 4 degrees C. Maximum encapsulation was observed at a ratio of 1:200 (Lipid:Halofantrine) Halofantrine is released from Halolipo at both pH 7.2 and 5.5. Halolipo causes decrease in cell viability of L. donovani Ag83 promastigotes and leading to cellular stress as evident from increased ROS generation and mitochondrial membrane depolarization. In this work, the sensitivity of the germination of different plant species (horticultural, grasses and spontaneous flora) was tested. The results indicate that most of the plants are sensitive to juglone. The comparison between naphthoquinones and commercial pesticides did not provide enough evidence to use naphthoquinones to control spontaneous flora. Assessing the toxicity of pesticides is crucial because the sustainability of the ecosystem could be endangered by their use. In this study, the toxicity of certain pesticides (metam Na, oxyfluorfen, glyphosate, metribuzin and diuron) was evaluated based on dehydrogenase activity (DHA). DHA has become a good bioindicator for assessing the toxicity of xenobiotics. A toxicity index was proposed to compare the different degrees of toxicity among the tested substances. Finally, a toxicity threshold was constructed using the slopes of the fitted lines of the data, assuming that the maximum toxicity (10) represented the slope of the biocide HgCl2 and that the minimum toxicity (1) was attributed to the DHA obtained from soil without pesticides. The Synthesis, Self-Assembled Structures, and Microbicidal Activity of Cationic Gemini Surfactants with Branched Tridecyl Chains

Cationic gemini surfactants with polymethylene spacer and linear alkyl chains containing an even number of carbon atoms have been extensively studied in the recent past, with the emphasis put on the determination of their aggregation behaviour in aqueous solution and their biological properties. However, the information on the aggregation of branched gemini surfactants with an odd number of carbon atoms in their alkyl chains is only sparsely reported in the literature. To help cover this gap in the research of cationic gemini surfactants, a series of branched bisammonium cationic gemini surfactants with an odd number of carbon atoms in alkyl chains (tridecane-2-yl chains) and a polymethylene spacer with a variable length ranging from 3 to 12 carbon atoms have been synthesized and investigated. Critical micelle concentration, which was determined by three methods, was found to be in the order 10(-4) mol/L. A comparison of the obtained data of the novel series of tridecyl chain geminis with those of gemini surfactants with dodecyl chains and an identical spacer structure revealed that structural differences between both series of gemini surfactants result in different aggregation and surface properties for surfactants with 6 and 8 methylene groups in the spacer (N,N’-bis(tridecane-2-yl)-N,N,N’,N’-tetramethylhexane-1,6-diaminium dibromide and N,N’-bis(tridecane-2-yl)-N,N,N’,N’-tetramethyloctane-1,8-diaminium dibromide) with the cmc values 8.2 x 10(-4) mol/L and 6.5 x 10(-4) mol/L, respectively, as determined by surface tension measurements. Particle size analysis showed the formation of small stable spherical micelles in the interval between 2.8 and 5 nm and with zeta potential around +50 mV, which are independent of surfactant concentration and increase with the increasing spacer length. Microbicidal activity of 13-s-13 gemini surfactants was found to be efficient against Gram-positive, Gram-negative bacteria and yeast.

Influence of the Surface Roughness of PEEK GRF30 and Ti6Al4V SLM on the Viability of Primary Human Osteoblasts Determined by the MTT Test

The aim of the study was to clearly determine whether selected modern medical materials and three dimensional printing allow for satisfactory viability of human osteoblasts, which is important from the point of view of the subsequent osseointegration process. Moreover, as implants are produced with various topography, the influence of surface roughness on viability of bone cells was evaluated. To conduct the research, primary human osteoblasts (PromoCell) were used. Cells were seeded on samples of glass-reinforced polyetheretherketone (30% of the filling), Ti6Al4V manufactured with the use of selective laser melting technology and forged Ti6Al4V with appropriately prepared variable surface roughness. To assess the viability of the tested cells the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide was used. Results showed that all evaluated materials do not exhibit cytotoxic properties. Moreover, on their basis it can be concluded that there is a certain surface topography (designated i.a. as roughness equal to approx. Ra = 0.30 mu m), which ensures the highest possible viability of human osteoblasts. On the basis of the received data, it can also be concluded that modern glass-reinforced polyetheretherketone or Ti6Al4V produced by rapid prototyping method allow to manufacture implants that should be effectively used in clinical conditions.

Recycling Lepidolite from Tantalum-Niobium Mine Tailings by a Combined Magnetic-Flotation Process Using a Novel Gemini Surfactant: From Tailings Dams to the Bling Raw Material of Lithium

On January 25, 2019, the tailings dam at Vale’s Corrego do Feijao mine (Brumadinho, Minas Gerais, Brazil) ruptured, releasing about 12 million cubic meters of tailings, killing over 240 people and posing a considerable and ongoing environmental hazard. Thus, the recovery of valuable minerals from tailings can contribute to a sustainable disposal of this hazardous residue, saving human lives and protecting the environment. The tailings of a tantalum-niobium mine usually contain a considerable amount of lepidolite resource. Nonetheless, the method of sustainably acquiring the lepidolite resource from tailings of a tantalum-niobium mine at low-temperature environments has always been needed. In this study, we synthesize a new amine-based gemini surfactant, butanediyl-alpha,omega-bis(dimethyldodecylammonium bromide) (BDB). It is introduced as a collector in the flotation of lepidolite from the tantalum-niobium mine tailings for the first time at low temperatures. We compare its capability to collect lepidolite with the monomeric surfactant 1-dodecylamine (DA). In bench-scale flotation experiments, the Li2O recovery of lepidolite concentrate with 80 g/t BDB was 10.96% and 31.85% higher than that with 160 g/t DA at 298 and 275 K, respectively, which indicated that BDB is a better collector for lepidolite than DA. BDB is a novel and superior collector for disposing of lepidolite tailings ponds.

Adenosine 5’-triphosphate (ATP), as a molecular currency for intracellular energy transfer, not only participates in various physiological activities, but also involves in various pathological processes of living cells. Therefore, the establishment of a simple, rapid, sensitive and accurate ATP analysis method is of great significance for the study of its physiological functions. In this work, we rationally design a near-infrared phenylboronic acid group-functionalized dicyanovinyl squaraine (SQ- PBA3), with which a supramolecular assembly was constructed for fluorescent imaging of ATP. In PBS buffer, SQ-PBA3 changes self-assembly when adding cetyltrimethyl ammonium bromide (CTAB) deriving fluorescence response of SQ-PBA3 to ATP with the maximum emission at 700 nm, due to the specificity of phenylboronic acid to diols and the multiple electrostatic interactions between SQ PBA3, ATP and CTAB molecules. Meanwhile, the probe was successfully applied to monitor intracellular ATP level in MCF-7 cells with high sensitivity and selectivity.
