Bhat, Sachin A.;Rao, D. S. Shankar;Prasad, S. Krishna;Yelamaggad, Channabasaveshwar V. research published 《 Chiral plasmonic liquid crystal gold nanoparticles: self-assembly into a circular dichroism responsive helical lamellar superstructure》, the research content is summarized as follows. Owing to their proven and promising potential in various technol. endeavors ranging from catalysis and sensing to invisibility cloaks made from metamaterials, chiral plasmonic superstructures resulting from the directed self-assembly of optically active metal nanoparticles (MNPs) have been pursued intensively in recent years. Several strategic efforts have emerged especially to accomplish advanced nanomaterials assembling into liquid crystalline (LC) helical structures, where MNPs are regularly packed in fluid/frozen arrays/layers or wires (columns). While the helical fluid columnar arrays (mol. wires) showing CD (CD) have been realized, the discovery of fluid chiral lamellar ordering, where the dielec. and conducting regimes are arranged alternatively, has hitherto remained highly elusive. Herein we report the first examples of monodisperse LC-gold NPs (LC-GNPs) self-assembling into a fluid/frozen lamellar structure exhibiting CD activity. Notably, these new, exceptional LC-GNPs have been realized by simple, hassle-free protocols that involve the room temperature addition of LC dimer-like arylamines to Au(III), where the amines not only reduce Au(III) to Au(0) but also bind strongly to the central GNP scaffold. Their mol. structure, mesomorphism, and ability to interact with circularly polarized light have been evidenced unambiguously and could play an important role in realizing metamaterials in the visible region.

Multiple Splice variants of AR have been reported in the past few years. These splice variants are upregulated in most cases of CRPC resulting in poor prognosis. Most of these variants lack the ligand binding domain (LBD) but still bind to DNA resulting in constitutive activation of downstream targets. The AR-V7 splice variant has been characterized extensively and current clin. trials in CRPC are exploring the use of AR-V7 as a biomarker. New therapeutic mols. that selectively target AR-V7 are also being explored. However, there is a dearth of information available on the selectivity, phenotypic responses in AR-V7 dependent cell lines and pharmacokinetic properties of such mols. Using our proprietary computational algorithms and rational SAR optimization, we have developed a potent and selective AR-V7 degrader from a known AR DNA binding domain (DBD) binder. This mol. effectively degraded AR-V7 in a CRPC cell line and demonstrated good oral bioavailability in mouse PK studies. This tool compound can be used to evaluate the pharmacol. effects of AR-V7 degraders. Further exploration of SAR can be pursued to develop more optimized lead compounds

Bai, Yulong;Wan, Wang;Huang, Yanan;Jin, Wenhan;Lyu, Haochen;Xia, Qiuxuan;Dong, Xuepeng;Gao, Zhenming;Liu, Yu research published 《 Quantitative interrogation of protein co-aggregation using multi-color fluorogenic protein aggregation sensors》, the research content is summarized as follows. Co-aggregation of multiple pathogenic proteins is common in neurodegenerative diseases but deconvolution of such biochem. process is challenging. Herein, we developed a dual-color fluorogenic thermal shift assay to simultaneously report on the aggregation of two different proteins and quant. study their thermodn. stability during co-aggregation. Expansion of spectral coverage was first achieved by developing multi-color fluorogenic protein aggregation sensors. Orthogonal detection was enabled by conjugating sensors of minimal fluorescence crosstalk to two different proteins via sortase-tag technol. Using this assay, we quantified shifts in melting temperatures in a heterozygous model protein system, revealing that the thermodn. stability of wild-type proteins was significantly compromised by the mutant ones but not vice versa. We also examined how small mol. ligands selectively and differentially interfere with such interplay. Finally, we demonstrated these sensors are suited to visualize how different proteins exert influence on each other upon their co-aggregation in live cells.

Targeting epigenetic dysregulation has emerged as a valuable therapeutic strategy in cancer treatment. Especially epigenetic combination therapy of histone deacetylase inhibitors (HDACi) with established anti-cancer drugs has provided promising results in preclin. and clin. studies. The structural optimization of alkoxyamide-based class I/IIb inhibitors afforded improved analogs with potent efficacy in cisplatin-resistant head and neck carcinoma cells and bortezomib-resistant leukemia cells. The most promising HDACi showed a superior synergistic cytotoxic activity as compared to vorinostat and class I HDACi in combination with cisplatin, leading to a full reversal of the chemoresistant phenotype in head and neck cancer cell lines, as well in combination with the proteasome inhibitors (bortezomib and carfilzomib) in a panel of leukemic cell lines. Furthermore, the most valuable alkoxyamide-based HDACi exhibited strong ex vivo anticancer efficacy against primary patient samples obtained from different therapy-resistant leukemic entities.
