Zhao, He;Wu, Yang;Ci, Chenggang;Tan, Zhenda;Yang, Jian;Jiang, Huanfeng;Dixneuf, Pierre H.;Zhang, Min research published 《 Intermolecular diastereoselective annulation of azaarenes into fused N-heterocycles by Ru(II) reductive catalysis》, the research content is summarized as follows. Derivatization of azaarenes can create mols. of biol. importance, but reductive functionalization of weakly reactive azaarenes remains a challenge. Here the authors show a dearomative, diastereoselective annulation of azaarenes, via ruthenium(II) reductive catalysis, proceeding with excellent selectivity, mild conditions, and broad substrate and functional group compatibility. Mechanistic studies reveal that the products are formed via hydride transfer-initiated β-aminomethylation and α-arylation of the pyridyl core in the azaarenes, and that paraformaldehyde serves as both the C1-building block and reductant precursor, and the use of Mg(OMe)2 base plays a critical role in determining the reaction chemoselectivity by lowering the hydrogen transfer rate. The present work opens a door to further develop valuable reductive functionalization of unsaturated systems by taking profit of formaldehyde-endowed two functions.

Zhang, Nan;Song, Di;Chen, Weijin;Zhang, Shenyan;Zhang, Panpan;Zhang, Na;Ma, Shutao research published 《 Modification of 5-methylphenanthridium from benzothiazoles to indoles as potent FtsZ inhibitors: Broadening the antibacterial spectrum toward vancomycin-resistant enterococci》, the research content is summarized as follows. The death caused by pathogenic bacteria has always been a severe threat to mankind. The prevalence of drug resistance among bacteria underscores an urgent goal for new antibacterial agents with novel mode of action. Here we first designed and synthesized a class of benzothiazolyl-5-methylphenanthridium derivatives and evaluated their antibacterial activity. On this basis, we further designed and synthesized another class of novel indolyl-5-methylphenanthridium derivatives by optimizing the benzothiazolyl-5-methylphenanthridium core and evaluated their antibacterial activity targeting the bacterial cell division protein FtsZ. The results showed that the indolyl-5-methylphenanthridium derivatives had greatly improved activity against various drug-resistant bacterial strains including methicillin-resistant Staphylococcus aureus and vancomycin-resistant Enterococcus (VRE). Among them, compound C5 displayed excellent antibacterial activity against susceptible (MIC = 1 μg/mL), methicillin-resistant and clin. isolated S. aureus (MIC = 2 μg/mL). With low hemolytic activity towards mice red blood cells, C5 exhibited good antibacterial effect in vivo in preliminary pharmacodynamic assay. More importantly, C5 was difficult to induce bacterial resistance. Further mechanism studies proved that C5 could inhibit bacterial cell division by promoting FtsZ polymerization, leading to disorderly polymerization and disordered knots. Therefore, our findings suggest that this class of novel indolyl-5-methylphenanthridium derivatives are promising for future antibacterial agents.

Twenty-nine 12 N-substituted aloperine derivatives were synthesized and screened for suppression on PD-L1 expression in H460 cells, as a continuation of our work. Systematic structural modifications led to the identification of compound I as the most active PD-L1 modulator. Compound I could significantly down-regulate both constitutive and inductive PD-L1 expression in NSCLC cells, and successively enhance the cytotoxicity of co-cultured T cells against tumor cells at the concentration of 20μM. Also, it exhibited a moderate in vivo anticancer efficacy against Lewis tumor xenograft with a stable PK and safety profile. The mechanism study indicated that I mediated the degradation of PD-L1 through a proteasome pathway, rather than a lysosome route. These results provided the powerful information for cancer immunotherapy of aloperine derivatives with unique endocyclic skeleton by targeting PD-L1 to activate immune cells to kill cancer cells.

A variety of minor organobromine compounds are found in nature, but none are biosynthesized or required by mammals. 402-49-3, formula is C8H6BrF3, Name is 1-(Bromomethyl)-4-(trifluoromethyl)benzene. Organobromine compounds have fallen under increased scrutiny for their environmental impact., Recommanded Product: 1-(Bromomethyl)-4-(trifluoromethyl)benzene.

Yuan, Yao;Zhang, Si-Yuan;Dong, Wu-Heng;Wu, Feng;Xie, Xiao-Min;Zhang, Zhao-Guo research published 《 Visible-Light-Induced Radical Cascade Cyclization of o-Diisocyanoarenes: Synthesis of Diethyl Benzo[a]phenazine-6,6(5H)-Dicarboxylate》, the research content is summarized as follows. A visible-light-induced radical cascade cyclization of ortho-diisocyanoarenes for the synthesis of di-Et benzo[a]phenazine-6,6(5H)-dicarboxylates has been developed. This process provides an efficient and convenient protocol for the construction of benzo[a]phenazine skeleton that widely present in biol. active mols. and pharmaceuticals. The reaction takes place under mild conditions and the products are obtained in moderate to excellent yields.

Recommanded Product: 1-(Bromomethyl)-4-(trifluoromethyl)benzene, 4-Trifluoromethylbenzyl bromide is a useful research compound. Its molecular formula is C8H6BrF3 and its molecular weight is 239.03 g/mol. The purity is usually 95%.
4-Trifluoromethylbenzyl bromide is a choline derivative that acts as an anticancer agent. It is structurally similar to the anticancer drug doxorubicin, which has been shown to be effective against breast cancer and leukemia. 4-Trifluoromethylbenzyl bromide interacts with cellular proteins, including choline kinase, and inhibits the mitochondrial pathway. This leads to cell death through apoptosis. The molecule also interacts with nucleotide bases such as thymine and cytosine in DNA, inhibiting transcription and replication. 4-Trifluoromethylbenzyl bromide binds strongly to the hydroxyl group of cholesterol by an electrophilic substitution mechanism to form a covalent bond with its hydroxy group. The molecule can also bind to chloride ions by an ionic bond., 402-49-3.
