Solvent-free and large area compatible deposition of methylammonium lead bromide perovskite by close space sublimation and its application in PIN diodes

In this work, a solvent-free method to deposit in-situ methylammonium lead bromide (MAPbBr(3)) perovskite thin films is discussed. Spin-coating methods used for hybrid perovskite deposition typically use solvents that might remain in the films after processing, potentially causing accelerated film degradation. Furthermore, spin-coating might not be suitable for applications that require thick films (>500 nm). To address these issues, a close space sublimation process was implemented to deposit solvent-free and in-situ MAPbBr(3) films with variable thicknesses. A post-deposition annealing protocol was developed that results in smooth perovskite films as thick as 3 mu m. Post-deposition anneal also reduces a photoluminescence emission band that originates from surface defects. The resulting films were chemically, optically and structurally studied and hysteresis-free PIN diodes with metal-oxide semiconductors as p and n-type layers were fabricated and analyzed. The results indicate that high quality perovskite films with variable thickness can be obtained by the close space sublimation (CSS) process demonstrated here. Although MAPbBr(3) was selected for this study, this CSS method demonstrated can be easily applied to other hybrid perovskites.

In vitro apoptotic effect on human lymphatic filarial parasite by piperidine derivatives and thymidine reversal study

A novel library of synthetic piperidine derivatives was used to screen against human lymphatic filarial parasite Brugia malayi. Piperidine has earlier been reported to have effect against parasites including rodent filarial nematodes. Compounds with hydroxyl substitutions (4Q and 4H) showed marked antifilarial effect. Molecular docking of 4H derivative showed more favorable thermodynamic parameters against thymidylate synthase of B. malayi than human counterpart. A wide difference between IC50 and LD50 ensured the therapeutic safety of the candidates against the filarial parasites. Addition of thymidine to the treatment regimen led to a significant reversal of antifilarial effect of 4H that confirmed inhibition of thymidylate synthase as pharmacological rationale. Apoptosis induced in the parasite as a consequence of probable inhibition of thymidylate synthase was studied by acridine orange/ethidium bromide fluorescent staining and poly (ADP-ribose) polymerase activity inhibition. Involvement of mitochondria was confirmed by decreased 3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide (MTT) conversion and increased cytosolic cytochrome c level in 4H treated microfilariae, compared with the untreated microfilariae. Moreover, Michael adduct of chalcone targeting dihydrofolate reductase and piperidine targeting thymidylate synthase demonstrated synergistic effect on the parasite, indicating the importance of inhibition of DNA synthesis by combined effect. In conclusion, piperidine derivatives with hydroxyl substitution have a great therapeutic potential with an apoptotic rationale involving mitochondrial pathway, due to possible inhibition of parasitic thymidylate synthase.

Cetyltriethylammonium bromide (CTEAB) was considered as a cationic surfactant to form lipoplexes with DNA. In TRIS/HCI buffer, CTEAB self-assembles above its critical micelle concentration (CMC = 0.15 mM) into small spherical micelles as determined by complementary scattering techniques. This surfactant readily interacts with the supercoiled plasmid DNA pBR322 via electrostatics and hydrophobic interactions. Upon increasing surfactant concentration, successive phase transitions are observed from partial neutralization to full compaction of DNA as evidenced by agarose gel electrophoresis, tensiometry, pyrene fluorescence, UV-Vis absorbance and circular dichroism measurements. Under UVA radiation (lambda >= 335 nm), we show that the presence of the surfactant increases photooxidized damage on DNA especially in the compacted state.
