Neuroprotective effects of mitoquinone and oleandrin on Parkinson’s disease model in zebrafish

Aim: The aim of this study is to investigate the possible protective effects of mitoquinone and oleandrin on rotenone induced Parkinson’s disease in zebrafish. Materials and methods: Adult zebrafish were exposed to rotenone and mitoquinone for 30 days. Biochemical parameters were determined by spectrophotometric method and Parkinson’s disease-related gene expressions were determined by reverse transcription polymerase chain reaction method. Measurement of neurotransmitters was performed by liquid chromatography tandem-mass spectrometry instrument. The accumulation of synuclein was demonstrated by immunohistochemical staining. In vitro thiazolyl blue tetrazolium bromide method was applied to determine the mitochondrial function of synaptosomal brain fractions using rotenone as a neurotoxic agent and mitoquinone and oleandrin as neuroprotective agents. Results: Mitoquinone improved the oxidant-antioxidant balance and neurotransmitter levels that were disrupted by rotenone. Mitoquinone also ameliorated the expressions of Parkinson’s disease-related gene expressions that were disrupted by rotenone. According to thiazolyl blue tetrazolium bromide assay results, mitoquinone and oleandrin increased mitochondrial function which was decreased due to rotenone exposure. Conclusion: Based on the results of our study, positive effects of mitoquinone were observed in Parkinson’s disease model induced by rotenone in zebrafish.

Biolayer interferometry provides a robust method for detecting DNA binding small molecules in microbial extracts

DNA replication is an exceptional point of therapeutic intervention for many cancer types and several small molecules targeting DNA have been developed into clinically used antitumor agents. Many of these molecules are naturally occurring metabolites from plants and microorganisms, such as the widely used chemotherapeutic doxorubicin. While natural product sources contain a vast number of DNA binding small molecules, isolating and identifying these molecules is challenging. Typical screening campaigns utilize time-consuming bioactivity-guided fractionation approaches, which use sequential rounds of cell-based assays to guide the isolation of active compounds. In this study, we explore the use of biolayer interferometry (BLI) as a tool for rapidly screening natural product sources for DNA targeting small molecules. We first verified that BLI robustly detected DNA binding using designed GC- and AT-rich DNA oligonucleotides with known DNA intercalating, groove binding, and covalent binding agents including actinomycin D (1), doxorubicin (2), ethidium bromide (3), propidium iodide (4), Hoechst 33342 (5), and netropsin (6). Although binding varied with the properties of the oligonucleotides, measured binding affinities agreed with previously reported values. We next utilized BLI to screen over 100 bacterial extracts from our microbial library for DNA binding activity and found three highly active extracts. Binding-guided isolation was used to isolate the active principle component from each extract, which were identified as echinomycin (8), actinomycin V (9), and chartreusin (10). This biosensor-based DNA binding screen is a novel, low-cost, easy to use, and sensitive approach for medium-throughput screening of complex chemical libraries.

Efficient electronic coupling and heterogeneous charge transport of zero-dimensional Cs4PbBr6 perovskite emitters

Organic and/or inorganic halide perovskites have become a new class of optoelectronic materials by virtue of excellent attractive physical and chemical characteristics. In this context, the zero-dimensional (0-D) cesium lead bromide (Cs4PbBr6) perovskite with outstanding photoluminescence (PL) properties has been recognized as the most promising candidate. However, the electron transfer dynamics and electrochemical behaviors of Cs4PbBr6 emitters still remain unclear. Herein, we prepared the 0-D Cs4PbBr6 perovskite emitters and investigated the exciton transport and electrochemical dynamics by utilizing temperature-dependent transient photoluminescence (TRPL) and electrogenerated chemiluminescence (ECL) techniques, respectively. Stronger electronic coupling of Cs4PbBr6 emitters arises from the overlap of electronic wavefunctions, indicating the increased possibility for the generation of electrochemiluminescence. Through the determination of the diffusion coefficient and electron-transfer rate by using an electrochemical method, we confirm that effective heterogeneous charge transfer at the interface of the electrode and electrolyte results in red-shifted ECL emission in the presence of benzoyl peroxide (BPO) as the co-reactant. The development of zero-dimensional perovskite emitters opens up a new avenue in the field of emerging optoelectronic and biosensing technologies, including but not limited to ECL devices, ECL immunoassays, organic light-emitting diodes, light-emitting electrochemical cells, and other bio-related detections.

A new class of mixed-valent europium halide ortho-oxoborates: Eu6X [BO3](4) (X = Cl and Br)

Their hexagonal crystal structures was determined by single-crystal X-ray diffraction and adopt the non-centrosymmetric space group P6(3)mc with lattice parameters in the range of a = 1052-1061 pm and c = 681-685 pm at 298 K as well as a = 1050-1059 pm and c = 680-682 pm at 100 K for Z = 2. Eight- and nine-fold coordinated europium cations have planar [BO3](3-) triangles and one of these also two halide anions as neighbors. The Raman and IR spectra of both compounds were acquired and compared to literature data to confirm the presence of isolated [BO3](3-) anions. DFT INVESTIGATION OF GEOMETRICAL STRUCTURE, IR AND RAMAN SPECTRA OF VINYL HALIDES CH2=CH-X (X IS F, Cl AND Br)

The geometrical structure and conformational energy stability of vinyl halides CH2=CH-X (X = F, CI, and Br) were examined by using Density Functional Theory calculations (B3LYP method in combination with 6-311G* basis sets). The values of HOMO-LUMO gap (Delta E) for vinyl halides (fluoride, chloride and bromide) were 7.68 (eV), 7.10 (eV) and 6.55 (eV) respectively. The calculated geometrical parameters were in good agreement with the previously observed results. Both HOMO-LUMO gap and geometrical parameters were found to account for the stability of the molecules. Electron distribution in HOMO-LUMO Frontier molecular orbitals was investigated to show the charge transfer within the molecules. The atomic charges and molecular electrostatic potential were interpreted together to demonstrate the electrophilic -nucleophilic reactivity. The vibrational frequencies were computed, the comparison between the three halides revealed that vinyl chloride and vinyl bromide have strong C-X stretching vibrations bands, but for vinyl fluoride the bands are weaker, the intensity characterization increasing from F to Br respectively.

Exploration of the soluting-out effect of carbohydrates on the micellization and surface activity of long-chain imidazolium ionic liquid in the aqueous medium

In order to elucidate the role of polyhydroxy compounds (PHC) in modifying the micellization and interfacial behaviour of surface-active ionic liquids (SAILs), system consisting of 1-tetradecyl-3-methylimidazolium bromide ([C(14)mim][Br]) in water and in aqueous solutions of D(+)-Xylose and D(+)-Glucose as additives at (298.15, 303.15 and 308.15) K using specific conductance, UV-visible spectroscopy, FT-IR spectroscopy, surface tension measurement was investigated. From the electrical conductivity measurements, various parameters such as critical micelle concentration (CMC), degree of ionization of the counterion on the micelles (alpha), the standard Gibbs energy of micellization (Delta G(m)(o)), etc. have been evaluated for different concentrations of carbohydrates at different temperatures. FT-IR spectroscopy is utilized to record the structural changes occurring in the studied systems at below CMC, at CMC, and above CMC concentrations. The various surface parameters including CMC, surface tension at CMC (gamma(cmc)), surface pressure at the interface (Pi(cmc)), the maximum surface excess concentration (Gamma(max)), and the minimum area per surfactant molecule at the surface, A(min) have also been obtained using surface tension measurements. The results obtained indicated that the CMC value of the IL in the presence of different concentrations of carbohydrates decreases in the order; D(+)-Glucose > D(+)-Xylose demonstrating that carbohydrates have a soluting-out effect and their tendency to lower the CMC value of IL increases with an increase of hydro-philicity of carbohydrates. A comprehensive study using conventional H-1, P-31 NMR as well as advanced PFGSE-DOSY NMR on structurally unique ionic liquids (ILs) having, in one hand, both phosphorus based counter ions called tetraalkylphosphonium diarylphosphate (PP)/diarylthiophosphate (PTP), and on the other hand polyisobutylenesuccinimide-tethered tetraalkylammonium diarylphosphate (PIBSI-P)/diarylthiophosphate (PIBSI-TP) has been carried out in order to establish their structures and interactions between the counter ions in chloroform solution. It has been noticed that the ion association increases significantly by increasing the cationic alkyl chain length in PP and moderately in PTP owing to stronger hydrophobic interactions. However in polymeric ionic liquids (POILs), PIBSI-P and PIBSI-TP, it is the hydrophobic effect of the long chain PIB backbone which dominates the course, effect of cationic alkyl chain length was not much observed. The 11s were synthesized either from an anion exchange reaction between tetraalkylphosphonium bromides and diaryiphosphate/thiophosphate (former) or from a reaction of same diarylphosphate/thiophosphate with tetraalkylammonium bromide derived from polyisobutylene succinic anhydride (PIBSA) (later).
