Low, Choon Heng;Buss, Joshua A.;Agapie, Theodor research published 《 Molybdenum-Mediated Coupling of Carbon Monoxide to a C3 Product on a Single Metal Site》, the research content is summarized as follows. The synthesis and characterization of naphthalenediyl-diphosphine Mo complexes are reported. A novel dicarbonyl-Mo complex 3 converts to bis(siloxy)acetylene complex 5 upon reduction and treatment with a silylelectrophile, Me3SiCl. This process shows exclusive C-C coupling distinct from the phenylene-linked analog that undergoes C-O cleavage. Further CO catenation can be engendered from 5 under mild conditions providing metallacyclobutenone complex 6, with a C3O3 organic motif derived from CO. Differences in reactivity are assigned to the nature of the arene linker, where the naphthalenediyl fragment shows a propensity for η4 binding previously not observed for phenylene. Consistent with this hypothesis, a Mo precursor with a 1,3-cyclohexadienediyl-based linker was prepared In agreement, this species also shows exclusive formation of a bis(siloxy)acetylene complex and subsequent coupling of a 3rd CO mol.

Liu, Haidong;Xing, Renyi;Ren, Kewei;Xue, Fei;Feng, Chao research published 《 α-Iminyl Cation-Involved Indole Construction via Bronsted Acid Promoted Reaction of Isoxazol-5-ones》, the research content is summarized as follows. Herein, a strategically novel method for the efficient construction of indole skeletons I (R1 = Ph, furan-2-yl, cyclohexyl, etc.; R2 = H, Me, C(O)OH; R1R2 = -((CH2)4)-; R3 = 5-Me, 7-Cl, 4-Br, etc.) using 2-phenylisoxazol-5-ones II (R4 = H, 4-Ph, 2-Me, 3,4-Cl2, etc.) as the starting material was reported. This reaction proceeds via Bronsted acid promoted α-iminyl cation generation by N-O bond cleavage and a subsequent intramol. cyclization to obtain 1H-indole-3-carboxylic acids III, which further undergoes decarboxylation to afford the final product. Control experiments show that the N-O bond cleavage and intramol. cyclization proceeds so fast that the 1H-indole-3-carboxylic acids III, could be isolated in high yields even after 5-10 min. The substrate scope of this transformation is broad and the desired products are obtained in moderate to good yields. The transition-metal-free reaction condition, CO2 as the sole byproduct, and good practicability adds synthetic potential of this transformation in pharmaceuticals and flavors industry.

To further explore the research of novel PARP-1 inhibitors, design and synthesis of a series of novel 4,5-dihydrospiro[benzo[c]azepine-1,1′-cyclohexan]-3(2H)-one derivatives I (R = H, 2-Cl, 3-F, etc.) PARP-1 inhibitors based on the author’s previous research is reported. Most compounds displayed certain antitumor activities against four tumor cell lines (A549, HepG2, HCT-116, and MCF-7). Specifically, the candidate compound I (R = 2-F) possessed strong anti-proliferative potency toward A549 cells with the IC50 value of 2.01μM and showed low toxicity to lung cancer cell line. And the in vitro enzyme inhibitory activity of compound I (R = 2-F) was better than rucaparib. Mol. docking studies provided a rational binding model of the above compound in complex with rucaparib. The following cell cycle and apoptosis assays revealed that this compound could arrest cell cycle in the S phase and induce cell apoptosis and western blot anal. further showed that it could effectively inhibit the PAR’s biosynthesis and was more effective than rucaparib. Overall, based on the biol. activity evaluation, compound I (R = 2-F) could be a potential lead compound for further developing novel amide PARP-1 inhibitors.

Li, Kai;Huang, Shengli;Liu, Tianyu;Jia, Shiqi;Yan, Hailong research published 《 Organocatalytic Asymmetric Dearomatizing Hetero-Diels-Alder Reaction of Nonactivated Arenes》, the research content is summarized as follows. Nonactivated arenes, such as benzene derivatives, are chem. inert due to their intrinsic aromaticity and low polarity. The catalytic asym. dearomatization (CADA, coined by You and co-workers) of the nonactivated arenes represents a formidable challenge. Herein, the authors demonstrate an organocatalytic asym. dearomatizing hetero-Diels-Alder reaction of benzene derivatives The tunable regioselectivity of this strategy allowed delivery of a diversity of stereochem. complex polycyclic compounds, e.g., I, and oxahelicenes, e.g., II, with excellent stereoselectivity. The high complexity and three-dimensionality of the products were crucial for their potential applications in materials science and drug discovery. Mechanistic studies suggested that this reaction proceeded through a chiral tetra-substituted vinylidene ortho-quinone methide (VQM) intermediate, which was extremely active to overcome the loss of aromaticity of benzene derivatives with concomitant chirality transfer.

Kurnia, Kiki Adi;Setyaningsih, Widiastuti;Darmawan, Noviyan;Yuliarto, Brian research published 《 A comprehensive study on the impact of the substituent on pKa of phenylboronic acid in aqueous and non-aqueous solutions: A computational approach》, the research content is summarized as follows. The acid dissociation constant (pKa) is the fundamental physicochem. properties required to understand the structure and reactivity of boronic acid-based material as a sensor that identifies carbohydrates. However, there is a lack of comprehensive study on the impact of the substituent on the pKa of monosubstituted phenylboronic acid in aqueous and non-aqueous solutions In this work, extensive exptl. data on the pKa of monosubstituted phenylboronic acid in an aqueous solution was reviewed and compared in terms of accuracy. In addition, computational, were used to predict and investigate the impact of the substituent on the pKa for a series of monosubstituted phenylboronic acid in an aqueous solution at the mol. level. Good agreement was observed between predicted and literature pKa values of monosubstituted phenylboronic acid in the aqueous solution While some deviations exist, predominantly with fluorine-containing phenylboronic acid, the COSMO-RS model is proficient at predicting the pKa of boronic acid in an aqueous solution with the accuracy of ±1.5 pKa. Subsequently, the model was used to predict the pKa of boronic acid in the non-aqueous solution, which data is not available in the literature. Furthermore, an excellent relationship is observed between the acidity of para-substituted, and to some extent, meta-substituted phenylboronic acid with the at. charge of acidic hydrogen calculated using Natural Bond Orbital (NBO) Population Anal. In contrast, the steric hindrance and the existence of other mol. forces might influence the acidity of ortho-substituted phenylboronic acid. The gathered information in this work could be of benefit for the understanding of the acidity of the boronic acid-based materials not only as a sensor but also in many diverse areas.

Accordingly, formal meta-selective transition-metal-free C-O and C-N couplings may be achieved. Mechanistic studies unambiguously support the aryne generation mechanism.

HPLC of Formula: 244205-40-1, 2-Bromophenylboronic Acid is used as an inhibitor of the hormone sensitive lipase.
2-Bromophenylboronic acid, also known as 2-Bromophenylboronic acid, is a useful research compound. Its molecular formula is C6H6BBrO2 and its molecular weight is 200.83 g/mol. The purity is usually 95%.
2-Bromophenylboronic acid is a glucose monitoring agent that has a ruthenium complex with an acidic environment. The nitro group and the amines are in close proximity to the boron center, and this proximity leads to a high nucleophilic character of the molecule. This reactivity allows 2-bromophenylboronic acid to be used as a fluorescence probe for acidic environments. 2-Bromophenylboronic acid also inhibits secretase enzymes, which are involved in Alzheimer’s disease and other neurodegenerative disorders. It is an inhibitor of γ-secretase, which is responsible for cleaving the amyloid precursor protein (APP), and it has shown efficacy against biphenyl, an anticancer drug that binds to benzodiazepine receptors. 2-Bromophenylboronic acid is also an enantiopure compound because all four substituents are different from each other., 244205-40-1.

Koszelewski, Dominik;Paprocki, Daniel;Brodzka, Anna;Keciek, Aleksandra;Wilk, Monika;Ostaszewski, Ryszard research published 《 The sustainable copper-catalyzed direct formation of highly functionalized p-quinols in water》, the research content is summarized as follows. The straightforward copper-catalyzed formation of p-quinols in water under air has been accomplished employing arylboronic acids. Various substituted p-benzoquinol derivatives of biol. importance were obtained in good yield. Furthermore, the synthesis of target p-quinols under physiol. conditions in serum and carbonate buffer is demonstrated. The essential features of this method are the high functional group tolerance and scalable one-pot preparation of p-quinols from corresponding arylboronic acid. Very mild reaction conditions, an absence of the heavy toxic metals, simple procedure as well as high chemoselectivity makes the established protocol desirable for academia and pharmaceutical industry laboratories

Kuleshova, Olena;Asako, Sobi;Ilies, Laurean research published 《 Ligand-Enabled, Iridium-Catalyzed ortho-Borylation of Fluoroarenes》, the research content is summarized as follows. A terpyridine derivative and an iridium complex catalyze the C-H borylation of a stoichiometric amount of a fluoroarene with high ortho-selectivity and tolerance of functional groups such as bromide, chloride, ester, ketone, amine, and in situ-borylated hydroxyl. Complex drug mols. such as haloperidol can be selectively borylated ortho to the F atom. The terpyridine ligand undergoes rollover cyclometalation to produce an N,N,C-coordinated iridium complex, which may either selectively borylate the fluoroarene by itself or undergo reductive elimination to produce a borylated ligand.
