Real-time PCR (qPCR) has been used for rapid identification of Salmonella Typhimurium and Staphylococcus aureus in dairy foods, but is unable to differentiate viable and unviable pathogens. Ethidium bromide monoazide (EMA), a DNA-intercalating agent, can detect only viable cells because selectively enter cells considered unviable and bind to their DNA, inhibiting its amplification during qPCR. The objective was to establish a protocol for detection of viable Salmonella Typhimurium and S. aureus, experimentally inoculated in coalho cheese, by the use of EMA combined with qPCR. The protocol was effective for the identification of viable Salmonella Typhimurium in coalho cheese but not for the S. aureus cells. Concentrations of viable Salmonella Typhimurium cells of 1 CFU/10 g of coalho cheese could be detected. The monoplex protocol enables the rapid and specific identification of viable Salmonella Typhimurium in coalho cheese, making it an alternative method for the quality and safety control of cheeses.

Biomagnification of ionizable organic compounds in rainbow trout Oncorhynchus mykiss

Background The assessment of persistence, bioaccumulation and toxicity (PBT) is part of the regulation process of ionic organic compounds (IOCs) and a major challenge, as a commonly acknowledged approach for the estimation of the bioaccumulation potential of IOCs is still missing. The goal of the present study was, therefore, to experimentally determine the bioaccumulation of fully ionized compounds and to identify screening parameters that can indicate high bioaccumulation potential of IOCs. Three feeding studies with rainbow trout (Oncorhynchus mykiss) were carried out according to OECD TG 305. Separation of liver, gastrointestinal tract (GIT) and carcass allowed to further elucidate the tissue distribution of the individual test substances. The chemicals chosen had characteristics that made them suspect for high bioaccumulation, and included two cations (tetrabutylphosphonium bromide (TBP), trimethyloctadecyl ammonium chloride (TMOA)) and four anions (benzotriazole, tecloftalam, pentachlorophenol (PCP), MEE-phosphonate). Data on the dietary biomagnification of IOCs (strong acids) were also collected from published literature. Results The highest distribution factors were found for the GIT, followed by liver. However, none of the tested IOCs showed a distinct biomagnification potential, as kinetic biomagnification factors (BMFk) ranged between 0.001 and 0.05 g/g (median 0.009 g/g). Cations showed lower assimilation efficiency (alpha) than anions, except for tecloftalam. In contrast, anions showed a considerably faster depuration rate (half-life less than 0.5 days) compared to cations (half-life of around 5 days). Sixteen potential screening parameters for BMF were calculated with a chemical property estimation tool (ACD/i-Lab) and correlated with the BMF data from this study and from literature. The number of hydrogen bond donors (nHBD) showed the highest correlation to measured BMF, but the prediction is only based on two values (one or two nHBD), while the other descriptors were insignificantly correlated. Conclusion The suspected dietary bioaccumulation potential of the six IOCs could not be confirmed in the feeding studies with rainbow trout. The more than twenty screening parameters showed no particularly high correlation neither with the test results nor with the BMF values collected from literature. The results corroborate earlier findings that ionization lowers the tendency of a chemical for dietary bioaccumulation, compared to non-ionized chemicals. In addition to the lipophobicity of ionic molecule moieties, fast depuration seems to be a major reason for the observed low dietary bioaccumulation of ionic compounds, in particular anions. Fast depuration may happen due to rapid metabolism of charged compounds, and future studies should test this hypothesis.

Introduction: Myasthenia gravis (MG) is an autoimmune disease in which antibodies directly target components of the neuromuscular junction, causing neuromuscular conduction damage that leads to muscle weakness. The current pharmaceutical treatment for MG is still not ideal to address the problems of disease progression, high recurrence rate, and drug side effects. Clinical observations suggest that traditional Chinese medicine (TCM) can strengthen immunity and improve symptoms of MG patients, delay the progression of the disease, reduce or even prevent the need for immunosuppressive therapy when used in combination with acetylcholinesterase inhibitors or low-dose prednisone, as well as improve the quality of life of patients. The Qiangji Jianli Capsule (QJC) is a combination of medicinal herbs which is used in traditional Chinese medicine. Since MG is a rare disorder, randomized controlled trials comparing large cohorts are difficult to conduct. Therefore, we proposed to aggregate data from a small series of N-of-1 trials to assess the effect of the Chinese medical prescription QJC, which strengthens the spleen and nourishes Qi, as an add-on treatment for MG with spleen and stomach Qi deficiency syndrome. Methods and analysis: Single-center, randomized, double-blind, multiple crossover N-of-1 studies will compare QJC versus placebo in 5 adult MG patients with spleen and stomach Qi deficiency syndrome. Patients will undergo 3 cycles of two 4-week intervention periods. According to the treatment schedule, patients will continue to be treated with pyridine bromide tablets, prednisone acetate, tablets and/or tacrolimus capsules throughout the entire trial. Each period consisting of 4-week oral add-on treatment with QJC will be compared with 4-week add-on treatment with a placebo. The primary endpoints are quantitative myasthenia gravis (QMG) test; measurement of the amount of T-reg cells and cytokines such as interferon-gamma (IFN-gamma), interleukin-4 (IL-4), interleukin-17A (IL-17A), and transforming growth factor-beta (TGF-beta); and corticosteroid or immunosuppressive agent dosage. Secondary outcome measures: Clinical: Evaluation of the effect of TCM syndromes; MG-activities of daily living (MG-ADL) scales; adverse events. Direct Z-Scheme Heterojunction of Semicoherent FAPbBr(3)/Bi2WO6 Interface for Photoredox Reaction with Large Driving Force

Metal halide perovskites with direct band gap and strong light absorption are promising materials for harvesting solar energy; however, their relatively narrow band gap limits their redox ability when used as a photocatalyst. Adding a second semiconductor component with the appropriate band structure offsets can generate a Z-scheme photocatalytic system, taking full advantage of the perovskite’s intrinsic properties. In this work, we develop a direct Z-scheme photocatalyst based on formamidinium lead bromide and bismuth tungstate (FAPbBr(3)/Bi2WO6) with strong redox ability for artificial solar-to-chemical energy conversion. With desirable band offsets and strong joint redox potential, the dual photocatalyst is shown to form a semicoherent heterointerface. Ultrafast transient infrared absorption studies employing selective excitation reveal synergetic photocarrier dynamics and demonstrate Z-scheme charge transfer mechanisms. Under simulated solar irradiation, a large driving force photoredox reaction (similar to 2.57 eV) of CO2 reduction coupled with benzyl alcohol oxidation to benzaldehyde is achieved on the Z-scheme FAPbBr(3)/Bi2WO6 photocatalyst, harnessing the full synergetic potential of the combined system.

Synthesis of hierarchical ZSM-5 aggregates by an alkali-treated seeds method with cetyltrimethylammonium bromide for the methanol to gasoline reaction

A series of hierarchical ZSM-5 aggregates with a relatively low SiO2/Al2O3 ratio (~ 50) were successfully synthesized using alkali-treated commercial ZSM-5 as seeds and cetyltrimethylammonium bromide (CTAB) as mesogenous template. The effect of the amounts of CTAB on the physicochemical and catalytic properties of the synthesized catalysts was characterized by XRD, FE-SEM, FE-TEM, N-2 physical adsorption, ICP-OES, NH3-TPD, Py-IR and TGA. Moreover, the possible formation mechanism of the hierarchical ZSM-5 aggregates was studied. The results indicated that the mesoporosity of the hierarchical ZSM-5 aggregates was greatly enhanced with the addition of CTAB. The primary crystal size of the ZSM-5 aggregates significantly decreased owing to the protective effect of CTAB, which inhibits the further crystal growth. The hierarchical ZSM-5 aggregates prepared with addition of a suitable amount of CTAB showed large specific surface areas and large external surface areas, abundant intercrystalline mesopores and appropriate concentration of acid sites, which resulted in excellent catalytic performance in the methanol to gasoline reaction. The stability of the catalyst was remarkably improved and the gasoline yield was given a rise of 10% compared with the catalyst prepared in the absence of CTAB.

Biosynthesis of gold nanoparticles using monosaccharides of Artemisia afra and their antioxidant and anticancer properties

Biogenic synthesis of gold nanoparticles has received much attention due to their varied medicinal application. Monosaccharides from Artemisia afra were used as reducing and capping agents for the synthesis of gold nanoparticles. The gold nanoparticles were characterised by ultraviolet-visible (UV-vis) spectroscopy, transmission electron microscopy (TEM) and scanning electron microscopy (SEM). The shape and morphology obtained for gold nanoparticles differed with different reaction conditions. TEM images of gold nanoparticles synthesised at high pH yielded a mixture of shapes (hexagons, rods and spheres) with average size of 7.75 nm whilst those synthesised at neutral pH yielded spheres with average size of 9.08 nm. Antioxidant activity of saccharides and gold nanoparticles by DPPH, ferric reducing antioxidant power and phosphomolybdenum methods showed gold nanoparticles synthesised at a high pH to have better antioxidant activity than those synthesised at neutral pH. The anticancer activity of the saccharides and gold nanoparticles was also investigated by the 3-[4,5-dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide (MTT) assay against the cancerous MCF-7 ( breast adenocarcinoma) and non-cancerous HEK293 (human embryonic kidney) cell lines. Gold nanoparticles exhibited excellent cytotoxic activity towards MCF-7 cells with those synthesised at neutral pH (compared to higher pH) showing greater cell viability towards HEK293.
